Lipopolysaccharide Reciprocally Alters the Stromal Cell-Regulated Positive and Negative Balance between Myelopoiesis and B Lymphopoiesis in C57BL/6 Mice

Abstract: Hematopoiesis in the bone marrow (BM) and spleen is controlled by stromal cells. Inflammation promotes myelopoiesis and simultaneously suppresses B lymphopoiesis. However, the role of the reciprocal regulation of myelopoiesis and B lymphopoiesis by stromal cells during inflammation is not fully understood. We investigated inflammation-induced alteration of hematopoietic regulation in lipopolysaccharide (LPS)-treated mice. C57BL/6 female mice were intravenously injected with a single, 5-µg dose of LPS, which in… Show more

“…In the same experiment, mRNA expression for various cytokines critical for maintenance of HSPCs was also evaluated ( Figure 2D). As previously reported, 21,22 LPS stimulation induced IL-6 and tumor necrosis factor (TNF)-a mRNA expression, but little or none of the other cytokine mRNAs assayed. In contrast, NOD1 stimulation under the same conditions induced IL-3, IL-7, Flt3L, SCF, and ThPO in addition to IL-6 and TNF-a mRNA expression.…”

“…6) However, the down-regulation of SDF-1, IL-7, and SCF in BALB/c mice treated with 5 µg LPS was more prolonged and profound than the decreases in C57BL/6 mice treated with 5 µg LPS. These different responses to LPS between BALB/c mice and C57BL/6 mice may be due to strain-dependent differences in stromal cell sensitivity to LPS and cytokines such as TNF-α, GM-CSF, and IL-6.…”

“…6,14,15) A control group of mice was treated with the same volume of pyrogen-free saline (0.2 mL per mouse). In the early-phase experiment, three mice from each treatment group were evaluated 1, 3, and 6 h and 1, 3, 5, and 7 d after the first, second, and third treatments with LPS or saline (Fig.…”
