Liquid-chromatographic measurement of biopterin and neopterin in serum and urine

Abstract: We report an improved "high-performance" liquid-chromatographic (HPLC) method for measuring biopterin and neopterin in serum and urine. Specimens are acidified, treated with iodine in 0.2 mol/L trichloroacetic acid, party purified on Bio-Rad MP-50 cation-exchange columns, and analyzed by reversed-phase HPLC with fluorometric detection. The minimal concentration of biopterin detectable is 0.3 micrograms/L in a 50-microL injection. The total CV is less than or equal to 10%. Improvements over other reported metho… Show more

“…10 Differential diagnosis is most commonly performed by analysis of urinary neopterin and biopterin. 11 Quantication of urinary neopterin and biopterin has been successfully performed by high-performance liquid chromatography (HPLC) coupled with electrochemical measurements, uorescence or MS. [12][13][14][15] Due to the complicated matrix, extensive sample pre-treatment, including extraction, pre-concentration and chromatographic separation, etc., which can take tens of minutes or even hours, is usually required. The cost of consumables, such as consumable kits for the extraction of samples, and solvents for sample extractions and chromatographic separations, is also relatively high.…”

“…Recently, a UPLC-MS/MS-based method has also been described by [124]. Analysis of pterin levels in human plasma and serum usually involves oxidation with either MnO 2 or differential oxidation with iodide/iodine after a filtration step [121]. Recently, a new protocol containing a derivatization step for the precise measurement of BH 4 has been published by [119].…”

“…The mean value obtained in healthy controls is 0.55 =k 0.14 IU mL-I (manufacturer's information). Determination of neopterin was by reversed-phase high-performance liquid chromatography with minor modifications [9]. Briefly, urine samples were centrifuged to remove debris, diluted in 1 to 10 with water containing dimethylpterine as an internal standard and injected directly onto a Techsphere 5 ODS column (HPLC Technology Ltd).…”
