Liver metastasis models of human colorectal carcinoma established in nude mice by orthotopic transplantation and their biologic characteristic

Abstract: AIM:To establish a liver metastasis model of human colorectal carcinoma in nude mice.METHODS:Orthotopic transplantation of histologically intact colorectal tissues from patients into colorectal mucosa of nude mice. Tumorgenicity, invasion, metastasis and morphological characteristics of the transplanted tumors were studied by light microscopy, electron microscopy and immunohistochemistry.RESULTS:Liver metastasis models of human colon carcinoma (HCA-HMN-1) and human rectal carcinoma (HRA-HMN-2) were established… Show more

“…The site of transplantation is usually between the serous layer and seromuscular layer of the greater curvature of stomach. The "suture method" [20] or the "gastric bursa method" [21][22] has many practical difficulties and disadvantag es in establishing the model of transplantation, for example, the tumor tissue is easy to fall off postoperatively; manipulation is complex and needs relatively high skills; and there may be great blood loss during suturing, and mortality is relatively high. OB glue is biologic glue and has been used widely in surgery owing to its secure wound adhesion.…”

“…With the combination of TRAIL and chemotherapy, the dose of chemotherapeutic agent could be reduced, and thus decreased the side effects of chemotherapy. In order to explore a more effective approach to deliver TRAIL gene for colorectal treatment, or to evaluate the effect of TRAIL gene on liver metastasis (through liver metastasis models of human colorectal carcinoma established in nude mice) [31] ,further research on it is necessary.…”
