In this study, fluorescence spectroscopy and molecular modeling approaches were employed to investigate the binding of methotrexate to human serum albumin (HSA) under physiological conditions. From the mechanism, it was demonstrated that fluorescence quenching of HSA by methotrexate results from the formation of a methotrexate/HSA complex. Binding parameters calculated using the Stern-Volmer method and the Scatchard method showed that methotrexate binds to HSA with binding affinities in the order 10 L·mol . Thermodynamic parameter studies revealed that the binding reaction is spontaneous, and that hydrogen bonds and van der Waals interactions play a major role in the reaction. Site marker competitive displacement experiments and a molecular modeling approach demonstrated that methotrexate binds with appropriate affinity to site I (subdomain IIA) of HSA. Furthermore, we discuss some factors that influence methotrexate binding to HSA.

The modified quantum dots (QDs) have been used in intracellular probing and drug delivery because of their special chemical and physical properties. In this paper, two β-cyclodextrin (β-CD)-modified CdSe/ZnS QDs with strong optical emission properties were synthesized as drug carriers to induce apoptosis. The positively charged l-Arginine (l-Arg) and neutral l-Tryptophan (l-Trp) were selected as ligands to compare the effect of charge on bioactivity of QDs nanoparticles. The in vitro assays revealed that these modified QDs showed good Dox carrier ability and significantly high inhibition rate to cancer cells. Especially, the more positively charged β-CD-l-Arg-polyamine-coated CdSe/ZnS QDs could effectively deliver the doxorubicin (Dox) into cells and exhibit excellent cell selectivity in cancer versus normal cells. The Dox-loaded QDs could enter intracellular, which showed that the Dox can efficiently go through the membranes at the existence of β-CD. Several lines of evidence suggest that the Dox-loaded QDs can efficiently induce apoptosis likely related to the production of ROS. We expect that the modified QDs can enhance the amount of hydrophobic antitumor drugs in cells and can also be used as fluorescent imaging agents.

The aim of this research is a comparison of the efficiency of six commercially available cyclodextrins (CDs) to improve the solubility and oral bioavailability of atorvastatin calcium (ATV‐Ca) and rosuvastatin calcium (ROV‐Ca) drugs in aqueous media. Inclusion complexes of both drugs with non‐toxic α‐CD, β‐CD, γ‐CD, HP‐β‐CD, M‐β‐CD, and maltodextrin were prepared in a 1:1 stoichiometry via the kneading method. To reach the best CD, various experimental and computational analyses were performed including phase solubility, dynamic light scattering (DLS), Fourier transform infrared spectroscopy (FT‐IR), X‐ray diffraction (XRD), differential scanning calorimetry (DSC), scanning electron microscopy (SEM), atomic force microscopy (AFM), hydrogen‐1 nuclear magnetic resonance (1HNMR), carbon‐13 nuclear magnetic resonance (13CNMR), and molecular docking calculations. The M‐β‐CD turned out to be the best substrate for the micro‐encapsulation of both drugs. Also, ATV showed a higher tendency than ROV to form inclusion complexes with CDs. Molecular docking studies showed that HP–β–CD and M‐β‐CD are the most suitable substrates for the formation of inclusion complexes, respectively. Our research showed that the β‐CD is not necessarily the most efficient substrate for increasing solubility based on previous reports in the literature; meanwhile, the other employed substrates in this study can show acceptable performances in this regard. According to our results, M‐β‐CD is the best substrate for the micro‐encapsulation of both drugs, which increases their solubility in water.
