SUMMARYAxonal transport of mitochondria was studied in frog sciatic nerves incubated in agents selected for their known or alleged effect on microtubules or axonal flow.Quantitative data on mitochondria, microtubules, neurofilaments, endoplasmic reticulum, and cross-sectional area of the axon indicate that axonal transport of mitochondria is dependent on microtubules. When more than half of the microtubules are destroyed, the axonal transport of mitochondria is diminished in proportion to the destruction of microtubules.Axonal transport of mitochondria is not related to neurofilaments and endoplasmic reticulum.Changes in the cross-sectional area of axons, even upon reduction to half the normal size, do not noticeably affect mitochondrial transport.Cyanide which blocks oxidative metabolism also blocks axonal transport of mitochondria, but analysis of fine structure indicates that cyanide is destructive to microtubules as well.

Cell functions such as adhesion, movement, and phagocytosis involve changes in both cell shape and cell surface motility. These changes are the result of membrane perturbations caused by interactions of cells with substrates, particles, or other cells. Surface perturbations generate signals that are presumably transduced into changes in the state of aggregation and distribution of submembranous cytoskeletal constituents (1-3). Except for the role of Fc and C3 receptors in the interaction of particles or substrates with phagocytes (3), other cell surface-"sensing" components, as well as the nature of the membrane perturbations and the transducing mechanisms are unknown. Induced cell shape changes permit the analysis of factors involved in maintenance of cell shape and in cell motility. A few diverse agents promote rapid and reversible changes in cell shape. Examples are the spreading of cells induced by Mn 2÷ (4) and the cell rounding induced by high hydrostatic pressure or exposure to trypsin, strychnine, EDTA, or cytochalasins (5-9). Cationic anesthetics inhibit a variety of cell surface phenomena, including cell to substrate adhesion, spreading, phagocytesis, locomotion, cell fusion, capping of surface Ig, or cell mediated-cytotoxicity (10-16). We show here that exposure of cultivated macrophages to cationic anesthetics results in extensive and fully reversible contraction and rounding. Single cells were followed before, during, and after exposure to the drugs and the observations recorded in still pictures and time-lapse movies. We examined several features of anesthetic-induced rounding, investigated structure-activity relationships of the drugs, and compared anesthetic effects to those of EDTA. Materials and Methods

Local anaesthetics are known to affect a variety of cell functions, many of which are involved in the inflammatory response. Local anaesthetics have also been shown to influence cell proliferation. The aim of this study was to examine the effect of two local anaesthetics (ropivacaine and lidocaine) on cell proliferation of cultured human fibroblasts, vascular endothelial cells and epithelial cells, i.e. keratinocytes, as earlier studies have not included primary human cell types. Significant inhibition of fibroblast proliferation was observed with concentrations of 50 microM ropivacaine or 100 microM lidocaine in 1% newborn calf serum and 500 microM ropivacaine or lidocaine in 10% newborn calf serum. The proliferation of endothelial cells was significantly inhibited by 1 mM ropivacaine in 5% human serum and 500 microM ropivacaine or 100 microM lidocaine in 40% human serum. Significant inhibition was not obtained with lidocaine when these cells were cultured in 5% HS. Significant inhibition of keratinocytes was obtained with 100 microM ropivacaine and 500 microM lidocaine. The effective concentrations are within the range of therapeutical concentrations in vivo and there seems to be a general correlation between the local anaesthetic potency and the inhibiting effect on cell proliferation. This suggest a mechanism by which local anaesthetics may exhibit anti-hyperproliferative effects in clinical situations.
