Localization and partial characterization of a rat ovarian granulose cell protein with a monoclonal antibody

Abstract: Hybridoma cell lines were obtained from mouse splenocytes sensitized to granulosa cells collected from rat ovaries after gonadotropin stimulation. A monoclonal antibody (5G5) was obtained which reacted with granulosa cells and showed a positive reaction with serum-free conditioned medium containing granulosa cell secreted proteins. Immunoblotting of the conditioned medium and light- and electron-microscopic immunocytochemistry of rat ovary show that mAb 5G5 is directed against a 59-kDa protein which is located… Show more

“…Further controls were performed with subclass-matched control Mab 5G5 (IgG) obtained against rat ovarian antigens, produced and characterized in our laboratory after immunization with granulosa cells and oocytes (Russinova et al, 1993(Russinova et al, , 1994). An additional control was performed with Mab 2F2 preabsorbed with protein extract obtained from the cells utilized for immunization before being processed for immunohistochemistry.…”

“…In controls, the primary antibody was replaced with (i) normal mouse serum; (ii) Mab 6C3 absorbed with synthetic ANF (20 µg ml -1 ) for 24 h at 4°C or (iii) PBS instead of Mab 6C3. Further controls were performed with Mab 5G5 (IgG) obtained against rat ovarian antigen which bound a 59-kDa protein located on granulosa and Leydig cell surface (Russinova et al, 1993;Russinova et al, 1995) as a subclass matched control.…”
