Localization of Multiple Quantitative Trait Loci Influencing Susceptibility to Infection withAscaris lumbricoides

Abstract: A linkage-based genome scan of 1,258 members of a single pedigree of the Jirel population of Nepal localized 6 potential quantitative trait loci (QTLs) influencing susceptibility to infection with Ascaris lumbricoides, the most common soil-transmitted intestinal helminth. Three QTLs exhibited genomewide significance, including QTLs on chromosomes 13 (logarithm of the odds ratio [LOD] score, 3.37; genomewide P = .013, 8 (LOD score, 3.03; genomewide P = .031), and 11 (LOD score, 3.19; genomewide P = .020). “…We also included four candidate genes in LD with SNP TXNIP (TNFAIP8L2, PIAS3, RFX5, RBM8A) (Skallerup et al 2012) as well as an additional candidate gene within our QTL (FCGR1A). In addition, we tested the expression of TNFSF13B, a candidate gene for A. lumbricoides burden (Williams-Blangero et al 2002;2008). A functional grouping of the genes is provided in Supplementary Table S1.…”

“…In addition to FCGR1A, we measured the expression of three candidate genes within our QTL (TNFAIP8L2, PIAS3, and RBM8A) as well as TNFSF13B, a candidate gene for A. lumbricoides burden (Williams-Blangero et al 2002;2008) located on porcine chromosome 11. We did not observe a significant change of expression levels in the two genotypes due to A. suum infection for any of these genes.…”

“…While the generative mechanisms of predisposition are currently unknown, it is believed that host genetics may play a major role [56][57][58][59][60][61]. Familial patterning to Ascaris infection has been M A N U S C R I P T indicates that the genetic factors influencing the host's capacity to respond protectively to infection are polygenic, specified by many minor susceptibility genes rather than primarily by a few major loci [62].…”

“…Detailed research has been conducted in East Nepal [56,60,61] using genome scanning of individual human hosts from a Jirel population (Jiri Helminth Project). The worm burden of the individuals in the study group was first assessed to determine the intensity level of infection.…”

“…These parasites include Ascaris lumbricoides , Cryptosporidium spp., and Entamoeba histolytica . [9][10][11][12][13][14] However, little is known about the importance of host genetics for risk of developing ECD. Knowledge of the genetic determinants of host susceptibility to ECD may facilitate development of novel approaches for control of the disease.…”
