Long-term culture of SH-SY5Y neuroblastoma cells in the absence of neurotrophins: A novel model of neuronal ageing

Abstract: Background Studying human ageing is of increasing importance due to the worldwide ageing population. However, it faces the challenge of lengthy experiments to produce an ageing phenotype. Often, to recreate the hallmarks of ageing requires complex empirical conditions that can confound data interpretation. Indeed, many studies use whole organisms with relatively short life spans, which may have little, or limited, relevance to human ageing. There has been extensive use of cell lines to study agein… Show more

“…Potential explanations for the observed lipid changes and potential beneficial properties of vitamin B12 in respect to AD are given in the Supplementary Discussion as further background information. However, these explanations are based on literature and should not be overinterpreted without further experimental proof [ 78 , 79 , 80 , 81 , 82 , 83 , 84 , 85 , 86 , 87 , 88 , 89 , 90 , 91 , 92 , 93 , 94 , 95 , 96 , 97 , 98 , 99 , 100 , 101 , 102 , 103 , 104 , 105 , 106 , 107 , 108 , 109 , 110 , 111 , 112 , 113 , 114 , 115 , 116 , 117 , 118 , 119 , 120 ].…”

“…We selected culture supernatants of hippocampal neurons cultured for 0, 7, and 14 days as the research object, which did not match the data source of biological analysis in a strict sense. We adopted this experimental design because we hoped to explore the changes in the intracellular environment specifically related to maturation. , Too short of a cell culture time will not be conducive to the analysis of components. The cells cultured for 0 days can be identified as the youngest hippocampal neurons, which will help us to explore the differential genes related to maturation in this study.…”

“…Several cell lines employed for anti-Alzheimer's candidate discovery are SH-SY5Y primary cell line, CHO cell line, human neuroblastoma SK-N-SH cell line, primary cultured rat microglia and peripheral blood mononuclear cell (PBMC) from AD patients. SH-SY5Y cell line SH-SY5Y primary culture cells are humanderived cells that have been widely used in various in vitro experiments, including for anti-Alzheimer's discovery (Strother et al, 2021). Since derived from human cells, SH-SY5Y cells express human-specific protein and protein isoforms.…”
