Long-term nebivolol administration reduces renal fibrosis and prevents endothelial dysfunction in rats with hypertension induced by renal mass reduction

Abstract: Nebivolol protects against renal fibrosis, oxidative stress and endothelial dysfunction better than equivalent doses, in terms of arterial pressure reduction, of atenolol in a hypertensive model of renal damage induced by RMR.

“…It leads to an increase in renal NO excretion and a significant increase in renal plasma flow and GFR, 37 reduces endothelin-1, 38 and has an antioxidant effect, 39 all of which are related to the pathogenesis of CN. Based on these characteristics of nebivolol, Toprak et al conducted an experimental study to investigate nebivolol's protective effects, 11 finding that nebivolol decreased medullary congestion, proteinaceous casts, and tubular necrosis caused by CM.…”

“…Hence, this model is used to study different aspects of CKD, namely to assess the potential renoprotective effects of new drugs (8,9). Detection of renal damage and fibrosis in this model is very important in chronic studies and finding a noninvasive means of its detection would provide tremendous utility in experimental studies.…”

“…RMR was performed in 17 rats. The animals were subjected to right nephrectomy and surgical removal of the left kidney poles (8). A sham operation was performed on 13 additional rats as a nonnephrectomized control (sham-operated; SO).…”

“…[9] It is reported to possess antihypertensive, anti-oxidant activity, and also reduces renal fibrosis and prevents endothelial dysfunction. [10,11] So far, no report has proved that treatment with nebivolol could improve the survival rate after renal warm I/R injury. The present study examined whether treatment with nebivolol improve the survival rate in a renal warm I/R injury using a rat model, as renal I/R injury is due to the over-activation of sympathetic β1 receptor activity that ultimately leads to a functional deficiency of NO.…”
