Loss of laminin and of the laminin receptor integrin subunit alpha 6 in situ correlates with cytokine induced down regulation of alpha 6 on fibroblast-like synoviocytes from rheumatoid arthritis

Abstract: Objective-To investigate in situ the expression of the integrin receptor subunits 6 and 1 and the distribution of the ligand laminin in the synovia from osteoarthritis (OA) and rheumatoid arthritis (RA) patients and to study the eVect of cytokines and antirheumatic drugs on the expression of the 6 and 1 integrin subunits on long term cultures of fibroblast-like synoviocytes (FBS) derived from OA and RA. Methods-The expression of the 6 and 1 integrin subunits and the distribution of laminin were examined immuno… Show more

“…This concept has been extended to disease-regulated therapeutic transgene expression (56 -58). Cytokines have been reported to downregulate ␣ v integrins in RA FLS and synovium (12,46). However, integrin expression levels do not correlate with Ad5-mediated transgene expression (13,59).…”

“…RA FLS and tissue are known to express multiple integrins (10). Among these, RGD-binding integrins such as ␣ v ␤ 3 , ␣ v ␤ 5 , ␣ 5 ␤ 1 , non-RGD-binding integrins such as ␣ 3 ␤ 1 , and LDV-binding integrins ␣ 4 ␤ 1 have been described (10,12,46). In murine synovial cells and tissue, CAR is also notably absent, and ␣ v ␤ 3 and ␣ v ␤ 5 integrins are expressed (13).…”

“…IL1-α and TNFα downregulate the expression of the α6 integrin in long-term cultures of synoviocytes obtained from patients with rheumatoid arthritis. Such changes in the expression of α6 occur concomitantly with the gradual loss of laminin 40. Both cytokines may be increased in some patients and promote downregulation of this integrin.…”

“…Remarkably, TNF-α repressed and MTX induced the expression of ITGA6 in long-term cultures of RASF. 29 Therefore, the loss of integrins in RASF may contribute to their evasion from inflamed synovial tissues prior to the formation of pannus tissue.…”
