Previously, we showed that cefotaxime (CTX) exposure increases conjugative transfer of a blaCTX–M–1 encoding IncI1 plasmid (IncI1/pST49/CTX-M-1) in Escherichia coli in a SOS-independent manner. This study aimed at investigating whether the observation was unique for that plasmid/strain/antibiotic combination or whether antibiotic-induced plasmid transfer (PT) is a more general phenomenon among plasmids in E. coli. Whole genome sequences of 25 E. coli strains were analyzed to identify different extended spectrum beta-lactamases (ESBL) plasmids enabling selection of a diverse collection of plasmids. Experiments were performed following exposure of these strains to 1/2 minimal inhibitory concentration (MIC) of CTX, ampicillin (AMP), or ciprofloxacin (CIP) before conjugation experiments. The frequency of PT was measured and compared to that of donors not exposed to antibiotics. Reverse-transcribed-quantitative real time polymerase chain reaction (RT-qPCR) was used to measure mRNA levels of five PT genes and two SOS response genes in donors exposed to antibiotics. The PT of eight strains (30.8% of strains tested) with IncI1/pST7/CTX-M-1, IncI1/pST49/CTX-M-1, IncI1/pST3/CTX-M-1, IncI1/pST293/CTX-M-1, IncI1/pST295/CTX-M-1, IncI1/pST16/CTX-M-55, and IncFII/CTX-M-14 (n = 2) plasmids was significantly increased following antibiotic exposure. CTX increased PT in all of these eight strain/plasmid combinations, AMP and CIP increased the PT in six and three strains, respectively. RT-qPCR showed that PT genes were up-regulated in the presence of the three antibiotics, whereas SOS-response genes were up-regulated only following CIP exposure. Our findings reveal that antibiotics can increase PT in E. coli strains with various ESBL plasmids. Thus, antibiotic-induced conjugative transfer of ESBL plasmids appears to be a common phenomenon in E. coli, having important implications for assessing the risks of antibiotic use.

The dissemination and propagation of antibiotic resistance genes (ARGs) via plasmid-mediated conjugation pose a major threat to global public health. The potential effects of nanomaterials on ARGs fates have drawn much attention recently. In this study, CeO 2 nanoparticles (NPs), one of the typical nanomaterials proposed for increasing crop production, were applied at the concentration range of 1−50 mg/L to investigate their effects on ARGs transfer between Escherichia coli. Our results revealed that the conjugative transfer of RP4 plasmid was enhanced by 118−123% at relatively high concentrations (25 and 50 mg/L) of CeO 2 NPs, however, CeO 2 NPs at low concentrations (1 and 5 mg/L) inhibited the transfer by 22−26%. The opposite effect at low concentrations is mainly attributed to (i) the reduced ROS level, (ii) the weakened intercellular contact via inhibiting the synthesis of polysaccharides in extracellular polymeric substances, and (iii) the down-regulated expression of plasmid transfer genes due to the shortage of ATP supply. Our findings highlight the distinct dose-dependent responses of ARGs conjugative transfer, providing evidence for selecting appropriate NPs dose to reduce the spread of ARGs while applying nanoagrotechnology.

Aims The aim of this study was to determine the effects of unsaturated fatty acids on clinical plasmids. Methods and Results Two unsaturated fatty acids, linoleic acid (LA) and α‐linolenic acid (ALA) at final concentration 0, 0·03, 0·3 and 3 mmol l−1, respectively, were used to assess the effects on conjugative transfer of a mcr‐1‐harbouring plasmid pCSZ4 (IncX4) in conjugation experiment. The inhibitory mechanisms were analysed by molecular docking and the gene expression of virB11 was quantitated by qRT‐PCR. Target plasmid diversity was carried out by TrwD/VirB11 homology protein sequence prediction analysis. Our results showed that LA and ALA inhibit plasmid pCSZ4 transfer by binding to the amino acid residues (Phe124 and Thr125) of VirB11 with dose‐dependent effects. The expression levels of virB11 gene were also significantly inhibited by LA and ALA treatment. Protein homology analysis revealed a wide distribution of TrwD/VirB11‐like genes among over 37 classes of plasmids originated from both Gram‐negative and Gram‐positive bacteria. Conclusions This study demonstrates representing a diversity of plasmids that may be potentially inhibited by unsaturated fatty acids. Significance and Impact of the Study Our work reported here provides additional support for application of curbing the spread of multiple plasmids by unsaturated fatty acids.
