Cerebrospinal Fluid Proteomics For Identification Of α2-Macroglobulin As A Potential Biomarker To Monitor Pharmacological Therapeutic Efficacy In Dopamine Dictated Disease States Of Parkinson’s Disease And Schizophrenia

Abstract: AimParkinson’s disease and schizophrenia are clinical end points of dopaminergic deficit and excess, respectively, in the mid-brain. In accordance, current pharmacological interventions aim to restore normal dopamine levels, the overshooting of which culminates in adverse effects which results in psychotic symptoms in Parkinson’s disease and extra-pyramidal symptoms in schizophrenia. Currently, there are no laboratory assays to assist treatment decisions or help foresee these drug side-effect outcomes. Therefo… Show more

“…Each experiment composed of at least one SLNM + , one SLNM- and, one of either of these two phenotypes or a cancer control benign breast tumor tissue. The equimolar culmination of the three phenotypes was made into an internal standard for the sake of normalization for each of the four experiments 27 . 80 μg of protein from each phenotype sample was reduced with 25 mM DTT for 30 min at 60 °C and alkylated with 55 mM iodoacetamide for 20 min at room temperature.…”

“…Moreover, the gene encoding this protein is considered as a PD risk gene [ 25 , 26 , 27 ]. It is noteworthy that in PD, the level of α-2-macroglobulin in the CSF changes, which is considered as a diagnostic marker [ 24 , 28 , 29 ]. Based on these data, one of the objectives of this study was to assess for the first time the activity of α-2-macroglobulin in the TF in PD patients.…”

“…In the recent past, our team has been actively involved in identifying potential protein biomarkers to understand various clinical phenotypes. [28][29][30] In this study, a 2-D fluorescence difference gel electrophoresis (DIGE) experiment has been carried out using sentinel lymph node tissue to identify a biomarker that can accurately reflect the metastatic lymph node in early breast cancer.…”
