<title>Three-dimensional reconstruction and analysis of gastric malignancies by electronic slides of consecutive sections and virtual microscopy</title>

Abstract: OBJECTIVE: The application ofan electronic slide and a software simulated virtual microscope can contribute to a more efficient, convenient histological analysis. These techniques would also support the automation of histological analysis and three dimensional reconstruction of histological objects. STUDY DESIGN: A fully computer controlled microscope (Axioplan 2 MOT), video camera(Grundig FAC 830) and an Intel Pentium II based PC were used for the development ofthe electronic slide. The applied frame grabber … Show more

“…For example, quantitative analysis of cells throughout an entire slide or for a specific region on a slide that harbors glands might help characterize different diseases. Ultimately, these algorithms should be applied to specimens that have been exhaustively sectioned and digitized so that entire specimens can be analyzed three-dimensionally (23)(24)(25). Therefore, our aim was to develop efficient algorithms to detect and measure higher order architecture and cytology using the recently developed Mirax Scan (Carl Zeiss Jena GmbH, Germany, Jena) digital slide scanner and determine whether these algorithms could be used by automated analysis to reliably diagnose gastric mucosa as normal or diseased on digital slides in the environment of virtual microscopy.…”

“…The very first publication about WSI, besides describing the technique itself, already applied this new technique to reconstruct gastric malignancies in 3D images. This work was published in 1999, years before other groups reintroduced WSI as a tool for 3D reconstruction [ 16 ]. In the recent years, with the improvement of WSI [ 17 ] and with the design of automated serial sectioning devices [ 18 ], more papers are being published in this field.…”

“…Recently, we have reported about a digital slide and virtual microscopy technique, the scanning fluorescent microscope (SFM) 12 . Digital slide means that mosaiced digital images of all fields of view on a part of the slide are made and matched by the computer to create a continuous image.…”
