Lyso-Lipid-Induced Oligodendrocyte Maturation Underlies Restoration of Optic Nerve Function

Abstract: Protein hyperdeimination and deficiency of lyso-phospholipids (LPC 18:1) has been associated with the pathology of demyelinating disease in both humans and mice. We uncovered interesting biology of LPC 18:1, in which LPC 18:1 induced optic nerve function restoration through oligodendrocyte maturation and remyelination in mouse model systems. Our in vitro studies show LPC 18:1 protection against neuron-ectopic hyperdeimination and stimulation of oligodendrocyte maturation, while … Show more

“…[77] Loss of oligodendrocytes causes demyelination and therefore impairs axon function and neuron, [78,79] while oligodendrocyte maturation promotes remyelination in mouse demyelination model. [80] In an MS mouse model, OPC differentiation is inhibited by both effector T cells and IFNš›¾ overexpression by astrocytes, and inhibiting immune activation of OPCs can facilitate remyelination. [81] A smaller fluorescent-positive area of CD4 + plus CD45 + , CD8 + plus CD45 + , and CD11b + plus CD45 + were found in the spinal cord with KD treated group, summarizing that fewer T cells are infiltrated into the diseased spinal cord, and KD treatment is capable of suppressing the infiltration of T cells.…”

“…Similarly, Lv et al 12 used a computational model for identifying drug candidates that were previously approved for other indications (nordihydroguaiaretic acid, vorinostat, and indomethacin) for breast and prostate cancers. Valdivia and Bhattacharya 8 implemented a similar genetic, proteomic, and lipidomic analysis approach to identify a lysolipid with the potential for the treatment of demyelinating diseases, such as multiple sclerosis and optic neuritis, attesting to the versatility of bioinformatics applications to different scientific fields. These studies have resulted in a significantly more efficient pathway to finding new drugs compared to the classic methods for treatment target identification.…”
