Biomaterial scaffolds mimicking the environment in metastatic organs can deconstruct complex signals and facilitate the study of cancer progression and metastasis. Here we report that a subcutaneous scaffold implant in mouse models of metastatic breast cancer in female mice recruits lung-tropic circulating tumor cells yet suppresses their growth through potent in situ antitumor immunity. In contrast, the lung, the endogenous metastatic organ for these models, develops lethal metastases in aggressive breast cancer, with less aggressive tumor models developing dormant lungs suppressing tumor growth. Our study reveals multifaceted roles of neutrophils in regulating metastasis. Breast cancer-educated neutrophils infiltrate the scaffold implants and lungs, secreting the same signal to attract lung-tropic circulating tumor cells. Second, antitumor and pro-tumor neutrophils are selectively recruited to the dormant scaffolds and lungs, respectively, responding to distinct groups of chemoattractants to establish activated or suppressive immune environments that direct different fates of cancer cells.

To investigate the efficacy of TP (Toripalimab Plus) chemotherapy combined with karelizumab in the treatment of advanced ovarian cancer after surgery. We recruited 96 patients with advanced ovarian cancer who were admitted to our hospital between December 2019 and December 2022 with advanced ovarian cancer and treated by tumor cell reduction surgery. The patients were randomly divided into groups using a digital table, 48 each. The control group was treated with chemotherapy, while the observation group was treated with chemotherapy combined with karelizumab. We compared the two groups with regards to adverse reactions and the levels of VEGF (Vascular Endothelial Growth Factor), MMP-9 (matrixmetalloproteinase 9), CEA (A Carcinoma Embryonic Antigen), AFP (alpha-fetoprotein), CA125 (carbohydrate antigen 125), CA19-9 (carbohydrate antigen 19-9) and T lymphocytes. There was no significant difference between the two groups with regards to adverse reactions (p > 0.05); the levels of VEGF and MMP-9 in the observation group were lower. There was a significant difference between the data before and after treatment within the same group (all p < 0.05); the levels of CEA, AFP, CA125 and CA19-9 in the observation group were lower than those in the control group, with significant differences before and after treatment within the same group (p < 0.05); the levels of CD3+ (cluster of differentiation 3+) and CD4+ in the observation group were higher, while the levels of CD8+ were lower than those in the control group. There were significant differences before and after treatment in the same group (p < 0.05). The combination of karelizumab and TP chemotherapy had a significant and positive impact on postoperative patients with advanced ovarian cancer by effectively regulating immune function and the levels of tumor markers. This protocol is safe and can be selected by considering the specific situation of individual patients.

Cancer continues to be a major global health issue causing millions of deaths annually. While traditional therapeutic methods may be effective in many cases, they may not be suitable for highly metastatic cancers. Moreover, the late detection of tumors, when they have already spread and are harder to treat, further exacerbates the challenge in managing this disease. As a result, there is a growing interest in developing complementary tissue‐engineered approaches for early cancer diagnosis and treatment to enhance patient recovery. Bioengineered cancer traps have gained significant attention due to their efficacy and ease of use. These trapping systems employ (bio)chemical and mechanical strategies to selectively capture and limit the spread of cancer cells, leading to their eradication from the body. Furthermore, when integrated into microfluidic devices, these cancer traps‐on‐a‐chip can be used for liquid biopsy and the early detection of circulating tumor cells and other tumor‐derived material, allowing for precision medicine treatments. Herein, this innovative approach to cancer theranostics, including its mechanism of action, current stage of development, and potential advantages and limitations is discussed.
