Macrophage Endocytosis of Superparamagnetic Iron Oxide Nanoparticles

Abstract: Competition experiments indicate that the cellular uptake of Ferumoxides involves scavenger receptor SR-A-mediated endocytosis. The comparison between Ferumoxides and Ferumoxtran-10 confirms that macrophage uptake of iron oxide nanoparticles depends mainly on the size of these contrast agents.

“…Ferumoxtran-10 (Sinerem, Guerbet, Aulnay-sous-Bois, France, and Combidex, Advanced Magnetics, Cambridge, Mass., USA) is a dextran-coated superparamagnetic iron oxide nanoparticle which is undergoing clinical phase III trials for the diagnosis of lymph node metastases [12]. In addition, it is also an appropriate MRI marker for the diagnosis of degenerative and inflammatory disorders, e.g.…”

“…Furthermore, the iron contained in ferumoxtran-10 was incorporated into the body's iron stock where increasingly found in the red blood cells as a constituent of hemoglobin structure [281]. As a result, the dextran-coated SPIONs did not show any chronic toxicity and their uptake by macrophages was not associated with cell activation [282][283][284].…”

“…NP uptake by leukocytes such as neutrophils and monocytes has been reported to occur mainly through the mechanism of phagocytosis, and to a small measure by pinocytosis. 22,23 On the contrary, lymphocytes do not possess the intrinsic ability of phagocytosis. 24 Lymphocytes internalize NPs by the mechanism of endocytosis.…”
