Magnetoliposomes: recent advances in the field of controlled drug delivery

Abstract: Magnetoliposomes have gained increasing attention as delivery systems, as they surpass many limitations associated with liposomes. The combination with magnetic nanoparticles provides a means for development of multimodal and multifunctional theranostic agents that enable on-demand drug release and real-time monitoring of therapy.Areas covered: Recently, several magnetoliposome structures have been reported to ensure efficient transport and delivery of therapeutics, while improving magnetic properties. Besides… Show more

“…Iron oxide-based nanoparticles, particularly magnetite, have attracted significant attention over the past two decades for developing biomedical applications mainly because iron is a common element that humans can metabolize 40 , 41 . In this regard, MNPs-based formulations have been approved by the Food and Drug Administration (FDA), mainly for magnetic resonance imaging purposes 42 , 43 and a few more for addressing Iron metabolic disorders such as Iron deficiency anemia 44 .…”

“…In contrast, Bacri et al showed that GVs containing superparamagnetic nanoparticles, that is, ferrofluid, are deformed by a weak magnetic field ( Figure 4 d) [ 110 ]. Liposomes encapsulating such nanoparticles are called magnetoliposomes and have drawn considerable attention as novel drug delivery carriers [ 111 ]. Nomura’s group reported that cell-sized magnetoliposomes can be remotely controlled and manipulated by an external magnetic field [ 112 ].…”

“…Given the stability of the liposomal formulation, slow action prevents complete drug delivery and creates the need to optimize active targeting by functionalization with various targeting ligands, such as proteins, peptides, nucleic acids, or small molecules. The targeting strategy involving mainly ligand-coupled liposomes is based on obtaining tumor-specific targeting through the interaction between the ligand and the receptor overexpressed in cancer cells [22][23][24][25]. The potential of liposomes for such modifications has led to various patents, which are listed in Table 1.…”
