MALDI Mass Spectral Imaging of Bile Acids Observed as Deprotonated Molecules and Proton-Bound Dimers from Mouse Liver Sections

Abstract: Bile acids (BAs) play two vital roles in living organisms, as they are involved in (1) the secretion of cholesterol from liver, and (2) the lipid digestion/absorption in the intestine. Abnormal bile acid synthesis or secretion can lead to severe liver disorders. Even though there is extensive literature on the mass spectrometric determination of BAs in biofluids and tissue homogenates, there are no reports on the spatial distribution in the biliary network of the liver. Here, we demonstrate the application of … Show more

For quantitative MALDI imaging of xylazine, a series of seven different calibration standard solutions was spotted onto the blank "unified brain" slices, followed by internal standard and DCTB matrix spraying.…”

“…Metabolite clusters were observed for the vasculature, hepatocytes, bile ducts, and the common bile duct. These clusters corresponded with co-registered ion images of heme B, a cofactor of hemoglobin that is enriched within the vasculature, and taurocholate, the most abundant bile acid (Fig 3D) (22, 23).…”

“…These were tentatively assigned based on accurate mass data to glyco (cheno)deoxycholate, taurocholate, and taurodeoxycholate, with further supportive MS/MS data in the last case. These compounds are naturally occurring bile salts that are normal constituents of liver tissue . Bile salt synthesis and secretion by the liver normally fluctuate with oral intake and are reduced during fasting .…”
