Malignant mast cell tumor of the thymus in an Royal College of Surgeons (RCS) rat

Abstract: A 152-week-old male Royal College of Surgeons (RCS) rat kept as a non-treated animal in a long-term animal study presented with a soft mass in the anterior mediastinum, which adhered to the pleura of the lung. Histopathologically, the mass mainly consisted of round to short spindle-shaped tumor cells that had infiltrated through the hyperplastic thymic tissue. The tumor cells were arranged in loose to dense sheets. Nuclei were moderate in size and round to spindle-shaped, with small nucleoli. Almost all tumor … Show more

“…82 Mast cell neoplasms are extremely rare and only isolated cases have been reported in rats. 99 In the NTP 2-year bioassays from 1982 to the present time containing 41 238 male and 43 612 female rats, only 4 instances of mast cell tumor have been reported in males and females (2 per sex). See “Incidences of Mast Cell Tumor and Histiocytic Sarcoma in Rats in NTP Studies” for more detailed data: NTP Incidence Data.…”

“…Number of animals examined for immunohistochemical analysis including normal thymuses, hyperplastic lesions, and thymomas are shown in Table1. The paraffin sections were analyzed immunohistochemically using antibodies against CK18 (Ks18.04, Mouse monoclonal, PROGEN Biotechnik, GmbH, Heidelberg, Germany) for cortical epithelium, 7 CK14 (EPR17350, Rabbit monoclonal; Abcam, Cambridge, MA, USA) for medullary epithelium, 9 and CD20 (M-20, Goat polyclonal; Santa Cruz Biotechnology, Inc., Santa Cruz, CA, USA) for B lymphocytes. Secondary antibody used the polymer kit (HISTOFINE Simple Stain Mouse MAX-PO; Nichirei Bioscience, Inc., Tokyo, Japan).…”

“…7,8 Additionally, Terayama et al reported that medullary thymic epithelial cells in the normal thymus of a Royal College of Surgeons rat are positive for CK14. 9 Brelińska et al reported that CK16 positive for thymic epithelial cells in the outer region of medulla of thymus in Wistar rats. 10 We focused on the differences in CK expression in the thymic epithelium of the cortex and medulla, investigated the type of thymic epithelium present in the cortex-like LyR, MD areas, and pale-staining cell foci observed in thymomas, and compared the structure of thymomas with the normal thymic cortex and medulla.…”

“…They reported that the granules partially showed a filament-like structure. In feline globule leukocyte tumor 14 or rat mast cell tumor 15 , however, granules contained nonstructural and homogenous materials. It was also reported that the characteristic feature of normal intestinal globule leukocytes in adult rats was the presence of paracrystalline inclusions in the granules 16 .…”

“…Globule leukocytes of the interepithelium of the gastrointestinal mucosa in rodents are positive for esterase 5 , 6 , mast cell protease 5 , 8 , 9 , 10 , granzyme B 9 , and c-kid 9 . Mast cells are also positive for esterase 5 , mast cell protease 5 , 9 , 15 , granzyme B 9 , and c-kid 9 , 15 , 17 . Granzyme B is a cytotoxic lymphocyte-associated protease in cytotoxic T cells or natural killer (NK) cells, although previous reports suggest that mast cells produce and release granzyme B upon activation 18 , 19 .…”
