Mammalian Phosphatidylinositol 4‐Kinases

Abstract: Three phosphatidylinositol 4-kinase isoforms, PI4K 230, 92 and 55 have been cloned and sequenced allowing a much wider characterization than the previously employed enzymological typing into type II and III enzymes. PI4K 230 and 92 contain a highly conserved catalytic core, PI4K55 one with a much lower degree of similarity. Candidate kinase motifs, deduced from the protein kinase super family, are absolutely conserved in all isoforms. Kinase activities are described based on their sensitivity and reactivity to… Show more

“…Consistent with a role for eEF1A2 in phospholipid signaling, we have previously reported that eEF1A2 binds to the lipid kinase phosphatidylinositol-4 kinase III beta (PI4KIII␤) and increases its kinase activity (21). PI4KIII␤ is a member of the PI4K family of lipid kinases that phosphorylates the D4 carbon of the inositol ring in phosphatidylinositol to yield phosphatidylinositol-4 phosphate (PI4P) (5,17). PI4Ks are emerging as important mediators of cell physiology because PI4P is itself a regulatory phospholipid and additionally an obligate precursor for PI(4,5)P 2 and PI(3,4,5)P 3 (4,29).…”

“…While the yeast orthologue of PI4KIII␣ is located at the plasma membrane (17), mammalian PI4KIII␣ mainly resides at the ER (18) but is also located at Golgi membranes (19) and at nucleoli upon exogenous expression (20). The localization of PI4KIII␣ also differs among cell types, since in neuronal cells and other cell types, like COS-7 or B50 cells, it is prominently found in the nuclei (21).…”

“…The first phosphorylation step is undertaken by phosphatidylinositol-4-kinase (PI4K) leading to the production of phosphatidylinositol-4 phosphate (PIP). Two groups of PI4 kinases, PI4K II and PI4K III, that show different sensitivity to the inhibitors Wortmannin, phenyl arsine oxide, and 5′-p-fluorosulfonylbenzoyladenosine (Barylko et al, 2002;Heilmeyer et al, 2003), are known to date. High levels of PI4K III mRNA can be found in various regions of the brain, among them the hippocampus (Zolyomi et al, 2000).…”
