MAPK signaling in neurodegeneration: influences of flavonoids and of nitric oxide

Abstract: Oxidative and nitrosative stress is increasingly associated with the pathology of neurodegeneration and aging. The molecular mechanisms underlying oxidative/nitrosative stress-induced neuronal damage are emerging and appear to involve a mode of death in which mitogen-activated protein kinase (MAPK) signaling pathways are strongly implicated. Thus, attention is turning towards the modulation of intracellular signaling as a therapeutic approach against neurodegeneration. Both endogenous and dietary agents have b… Show more

“…102,106 The action of flavonoids on the ERK pathway 83,99,107−110 appears to be mediated by interactions with MAPK kinases MEK1 and MEK2 and potentially membrane receptors. 102,111,112 In fact, flavonoids have close structural homology to specific pharmacological modulators of ERK signaling such as PD98059 (2′-amino-3′-methoxyflavone). Moreover, activation of ERK can result in downstream activation of cAMP response element binding protein (CREB), which may lead to changes in synaptic plasticity and memory 113,114 and to upregulation of neuroprotective pathways.…”

“…They may interact with specific proteins involved in intracellular signalling pathways (39) . They can selectively act on protein kinase and lipid kinase signalling cascades such as MAPK, protein kinase C, protein kinase B (PKB/AKT) (serine/threonine kinase), tyrosine kinases and phosphoinositide 3-kinase (PI3K) (for a review, see Williams et al (40) ).…”

“…However, it has been speculated that their classical hydrogen donating antioxidant activity is not the explanation for the bioactivity of flavonoids in vivo. Indeed, it has become evident that flavonoids are more likely to exert their neuroprotective actions by (1) the modulation of intracellular signalling cascades which control neuronal survival, death and differentiation; (2) affecting gene expression and (3) interactions with mitochondria [148,159,185]. This review will highlight some of the interactions of flavonoids with major neuronal intracellular signalling pathways, in particular those that are vital in determining neuronal death, survival, differentiation and proliferation.…”
