Abstract. Aerobic anoxygenic phototrophic (AAP) bacteria play significant roles in the bacterioplankton productivity and biogeochemical cycles of the surface ocean. In this study, we applied both cultivation and mRNA-based molecular methods to explore the diversity of AAP bacteria along an oligotrophic gradient in the Mediterranean Sea in early summer 2008. Colony-forming units obtained on three different agar media were screened for the production of bacteriochlorophyll-a (BChl-a), the light-harvesting pigment of AAP bacteria. BChl-a-containing colonies represented a low part of the cultivable fraction. In total, 54 AAP strains were isolated and the phylogenetic analyses based on their 16S rRNA and pufM genes showed that they were all affiliated to the Alphaproteobacteria. The most frequently isolated strains belonged to Citromicrobium bathyomarinum, and Erythrobacter and Roseovarius species. Most other isolates were related to species not reported to produce BChl-a and/or may represent novel taxa. Direct extraction of RNA from seawater samples enabled the analysis of the expression of pufM, the gene coding for the M subunit of the reaction centre complex of aerobic anoxygenic photosynthesis. Clone libraries of pufM gene transcripts revealed that most phylotypes were highly similar to sequences previously recovered from the Mediterranean Sea and a large majority (∼94 %) was affiliated to the Gammaproteobacteria. The most abundantly detected phylotypes occurred in the western and eastern Mediterranean basins. However, some were exclusivelyCorrespondence to: C. Jeanthon (jeanthon@sb-roscoff.fr) detected in the eastern basin, reflecting the highest diversity of pufM transcripts observed in this ultra-oligotrophic region. To our knowledge, this is the first study to document extensively the diversity of AAP isolates and to unveil the active AAP community in an oligotrophic marine environment. By pointing out the discrepancies between culture-based and molecular methods, this study highlights the existing gaps in the understanding of the AAP bacteria ecology, especially in the Mediterranean Sea and likely globally.

A Gram-negative-staining, non-motile, non-spore-forming, aerobic and rod-shaped bacterial strain, DPG-138 T , was isolated from seawater on the southern coast in Korea. Strain DPG-138 T grew optimally at 30 6C and in the presence of 2-3 % (w/v) NaCl. A neighbour-joining phylogenetic tree based on 16S rRNA gene sequences revealed that strain DPG-138T fell within the cluster comprising the type strains of four species of the genus Marivita (sequence similarity values of 97.8-98.2 %). The DNA G+C content was 59.9 mol%. The predominant ubiquinone of strain DPG-138 T was Q-10, typical for the genus Marivita.

A Gram-stain-negative, aerobic, non-motile and coccoid, ovoid or rod-shaped bacterial strain, BS-B2T, which was isolated from a tidal flat sediment at Boseong in South Korea, was characterized taxonomically. Strain BS-B2T grew optimally at 30 °C, at pH 7.0–8.0 and in the presence of 2.0 % (w/v) NaCl. The novel strain exhibited highest 16S rRNA gene sequence similarity (97.4 %) to Marivita geojedonensis DPG-138T. Neighbour-joining, maximum-likelihood and maximum-parsimony phylogenetic trees based on 16S rRNA gene sequences revealed that strain BS-B2T is closely related to Primorskyibacter sedentarius KMM 9018T, showing 96.5 % sequence similarity. Strain BS-B2T contained Q-10 as the predominant ubiquinone and C18 : 1ω7c as the predominant fatty acid. The polar lipid profile of strain BS-B2T comprised phosphatidylcholine, phosphatidylglycerol, one unidentified aminolipid and one unidentified lipid as major components, and differentiated it from the type strains of P. sedentarius and M. geojedonensis . The DNA G+C content of strain BS-B2T was 62.2 mol%. Differential phenotypic properties, together with the phylogenetic and chemotaxonomic data, demonstrated that strain BS-B2T can be distinguished from phylogenetically related genera as well as P. sedentarius and M. geojedonensis . On the basis of the data presented, strain BS-B2T is considered to represent a novel species of a new genus, for which the name Aestuariivita boseongensis gen. nov., sp. nov. is proposed. The type strain of Aestuariivita boseongensis is BS-B2T ( = KCTC 42052T = CECT 8532T).
