Matrine Attenuates D‐Galactose‐Induced Aging‐Related Behavior in Mice via Inhibition of Cellular Senescence and Oxidative Stress

Abstract: The present study was designed to evaluate the effects of matrine (MAT) on D-galactose- (D-gal-) induced aging and relative mechanism. Vitamin E at the dose of 100 mg/kg was used as a standard positive control. MAT significantly improved the D-gal-induced recognition and spatial memory impairment in novel object recognition and Y maze tests, and exercise endurance decreased in the weight-loaded swimming test at 2 and 10 mg/kg. We found that D-gal treatment induced noticeably aging-related changes such as reduc… Show more

“…Oxidative stress promoted cellular senescence [35]. A large number of studies have used the D-gal-induced ageing model for screening of anti-ageing compounds [26][27][28][29][32][33][34]. In our study, the D-gal-induced ageing model was well-established…”

“…d-Gal can cause systematic ageing-related changes, such as hepatic and brain injury, a decline in immune function [26][27][28][29], learning and memory impairment, and fatigue in behavior studies [26,27]. Parallel histopathological features of the liver, spleen, and hippocampus were observed similarly in the d-gal-induced ageing and naturally aging groups [30].…”

“…Meanwhile, MT suppressed the activation of p19/p21 and p16 pathways in the liver and hippocampus of D-gal-induced mice. These results suggest that MT plays an anti-aging role by inhibiting oxidative stress and cell senescence ( Sun et al., 2018 ).…”

“…5,6 It causes changes similar to natural aging, including biochemical changes, low immune function, cell growth reduction, decreased reproductive capacity and thymus coefficient. 7 Resveratrol (3,4',5-trans-trihydroxystilbene, RSV), an antioxidant, exists in numerous plants and red wine. Previous studies have shown that RSV has an antioxidant and anti-inflammatory effect.…”
