Matrix Metalloproteinases and Tissue lnhibitors of Matrix Metalloproteinases in Sputum from Patients with Bronchial Asthma

Abstract: To examine a possibility that matrix metalloproteinases (MMPs) participate in the pathogenesis of asthma and/or the development of asthma attack, we measured the concentrations of MMP-2, MMP-9, and their respective tissue inhibitors of metalloproteinases (TIMP)-2 and TIMP-1, in induced sputa collected from 28 patients with moderate to severe bronchial asthma. Specimens were collected during both the attack and the remission from 15 age- and sex-matched healthy control subjects. The concentration of MMP-9 was s… Show more

“…MMP-2 is activated at the cell surface by a trimolecular complex with TIMP-2 and the membrane-associated MMP-14 (5). The MMPs and TIMPs are being increasingly linked to asthma (29): there is increased expression of MMP-9, -2, and -3 in the airways of patients with asthma (35,48), MMP-9 is increased in lavage fluid during asthma exacerbations and allergen challenge (28) and is reduced when asthma control improves (21). In animal models of asthma, MMPs have major roles in migration of inflammatory cells through the airway, although this is complex.…”

“…[8,44] IL1R1 is also implicated in inflammatory diseases such as osteoarthritis [45] and multiple sclerosis. [46] It is important to note that the soluble form of IL1R1 is known to be anti-inflammatory and immunosuppressive through the sequestration of ILL [47] As asthma is an inflammatory Our study shows that 14 SNPs located in seven genes belonging to the IL1R2 gene activities are associated with at least one asthma-related phenotype in at least one population, which is an important finding. Except for IL1R1, which is in the 2ql2.1 region that contains the IL1R2, IL1RL2 and IL18RAP genes and that was already associated with asthma, [9] this is the first time that significant association with an asthma-related phenotype is reported for BACE1, SPI1, ERAP1, and IL 1 RAP.…”

“…MMP-9 was increased in both atopic and nonatopic wheezers and we consider this to be a marker for acute inflammation, as adult studies show that MMP-9 increases during exacerbations of asthma (24,26,27,31,32). In our study TIMP-1 excess was the rule as MMP-9 only exceeded TIMP-1 molar concentrations in one child, who clearly had a lower respiratory tract infection; yet raised MMP-9 was also seen in those with positive bacterial culture of BAL.…”
