Measurement of the Direct Oral Anticoagulants Apixaban, Dabigatran, Edoxaban, and Rivaroxaban in Human Plasma Using Turbulent Flow Liquid Chromatography With High-Resolution Mass Spectrometry

Abstract: The method is suitable for high-throughput therapeutic drug monitoring of DOACs. The acquisition of full scan data allows for the retrospective identification of metabolites. The method can be used to identify a particular DOAC if information on the drug taken is lacking.

“…Next steps for this transition will include the need for high mass accuracy instrumentation to overcome interferences that can affect the accuracy of measurement with 14 some nominal mass MRM systems [18,[26][27]. Initial exploration of this technology has already been done in toxicology laboratories [28]. It is envisaged that, with the perpetuating interest in the gut microbiome and health, research interest in TMAO and other gut-related analytes will increase (e.g.…”

“…Our developed and validate method is very simple and rapid, requires a smaller plasma volume (50 μL) and a shorter time of analysis than other methods reported in the scientific literature [18,20,21,24]. The instrumental analysis is performed in 1 minute and a set of 24 samples requires a preparation time of about 15 minutes.…”

“…The range of calibration was 1-500 ng/ mL, but our clinical practice demonstrated higher plasma levels of apixaban in patients after repeated administration than this range of concentrations (Fig.4). Processed volume plasma was 100 μL and total time of analysis was 6 min [18]. Schmitz et al quantified dabigatran, rivaroxaban and apixaban in plasma by ultra performance liquid chromatography (UPLC) coupled with tandem MS/MS as reference method to evaluate the analytical performance of several coagulation assays.…”

“…The literature is poor regarding the reports on the assay of EXN. Bathala et al, 6 analyzed EXN and its four metabolites in human plasma, urine and fecal samples, after oral administration of [ 14 C] EXN to 6 healthy male subjects, by either high-performance liquid chromatography/tandem mass spectrometry or a liquid chromatography radiometric method. This study was used to determine the mass balance and pharmacokinetics of EXN in humans.…”
