Measurement of the Subjective Effects of Methylphenidate in 11- to 15-Year-Old Children with Attention-Deficit/Hyperactivity Disorder

Abstract: Objective-This study examined subjective and other behavioral effects of methylphenidate (MPH) among adolescents.Methods-Standard abuse liability assessment methods that have been used in adult populations were modified for attention-deficit/hyperactivity disorder (ADHD) adolescents. MPH effects (0, 0.25 mg/kg) were evaluated under randomized, double-blind conditions in two 5-hour laboratory sessions in 24 (13 female) 11-15 year olds diagnosed with ADHD.

“…The peak plasma concentration of methylphenidate-IR is reached approximately 1–2 h after ingestion and the half-life of the drug in plasma is 2–4 h (Gualtieri et al, 1982). Earlier literature has revealed that peak effects from methylphenidate-IR on the performance of the CCPT are observed 1–2 h after it is taken in children and adolescents with ADHD (Martin et al, 2007). Therefore, the CCPT was administered 90 min after ingestion of the drug to maximize responses during the test session (Gualtieri et al, 1982).…”

“…Addiction Research Center Inventory. Participants completed the short form of the ARCI, which is a 48-item questionnaire, adapted by Martin et al for use in developing children, 34 and consists of five subscales: amphetamine (A), a scale that provides an assessment of amphetamine-like effects, including ''I have a weird feeling'' or ''I feel more excited than dreamy''; benzedrine group (BG), a scale that provides a measure of benzedrinelike effects, intellectual efficiency, and energy, including ''My movements seem slower today'' and ''People might say I am a little too dull today''; lysergic acid diethylamide (LSD), a scale that provides a measure of dysphoria and somatic complaints, including ''I would be happy all the time if I felt as I do now'' and ''My movements seem faster than usual''; morphine-benzedrine group (MBG), a scale that provides a measure of euphoria, including ''Things around me seem more pleasing than usual'' and ''I feel so good that I know other people can tell it''; and pentobarbital-chlorpromazine-alcohol group (PCAG), a scale that provides a measure of sedation, including ''I feel drowsy'' and ''It seems harder to move around today''. 35 Subjects completed the ARCI once before and *30 and 60 minutes after beverage consumption.…”
