Measurements of absolute values in biochemical fluorescence spectroscopy

Abstract: Fluoresce nce spectroscopy is an important tool of the bioc he mist studyin g th e structure and interac tion s of prote in s and nu cle ic acid s. The four bas ic quantities to measure accurately are: 1. s pec tra (co rrected excitation and e mi ss ion). 2. quantum yields (0), 3. fluoresce nce decay c haracte ri stics, and 4 _ poLarization_ Co mmercially avail ab le instruments, with little modification, can be used to obtain these meas ure me nts, but th e bioc he mi st in this fi eld is ve ry de pende nt on… Show more

“…The emission spectra on a wavenumber or energy scale were obtained by multiplying the measured and corrected emission spectra on a wavelength scale with l 2 [61]. The f f values of the DY series, ICG, and FabAntiCEAdDY-676 were calculated from integrated, blank, and spectrally corrected emission spectra (wavelength scale; prior to integration multiplication with l) relative to the standards IR 125 and Oxazine 1 using the following formula [62,63]:…”

“…7c shows the observed tryptophan spectrum after the signal processing, superimposed with the reference tryptophan spectrum (from ref. 22). The characteristic broad peak of tryptophan fluorescence is clear.…”

“…In addition, there exists extensive literature on potential fluorescence standards. This includes for example emission and excitation standards and standards for fluorometric quantities like fluorescence quantum yield, fluorescence lifetime as well as fluorescence (de)polarization [1][2][3][4][14][15]51,52].…”
