Measuring Uncertainty Using Different Approaches: A Case Study for Acetaminophen Quantification

Abstract: Investigation of out-of-specification analytical results is laborious, time-consuming, and costly and must be well documented. However, an analytical result is not complete unless reported with its measurement uncertainty. Here, we compare four different approaches for measurement uncertainty estimation used in acetaminophen quantification in pharmaceutical drug products. Measurement uncertainties were estimated using a repeatability and reproducibility study, Eurachem/Citac guidelines, Monte Carlo simulations… Show more

“…Measurement uncertainty estimation would be easier using other approaches such as method validation and repeatability/reproducibility studies. However, these approaches do not provide information of individual contributions of each source of uncertainty (Diaconu et al, 2015;Francisco, Saviano, Lourenço, 2016;Takano et al, 2017). Thus, we estimated the measurement uncertainty by Monte Carlo simulations performed using the regression equation of the amount of acetaminophen dissolved (%) as a function of time of dissolution (A), volume of dissolution media (B), pH of dissolution media (C), and rotation speed (D) ( Table II).…”
