Mechanism of anticancer action of novel berenil complex of platinum(II) combined with anti-MUC1 in MCF-7 breast cancer cells

Abstract: Mucin 1 (MUC1) is a high molecular weight transmembrane glycoprotein, that is overexpressed in >90% of breast cancers. It serves a crucial role in anti-apoptosis and tumor progression. MUC1 interacts with proteins in the extracellular matrix, at the cell membrane, in the cytoplasm and in the nucleus. The aim of the present study was to investigate the mechanism of anticancer action induced by novel berenil complex of platinum(II) (Pt12) together with a monoclonal antibody against MUC1 in breast cancer MCF-7 ce… Show more

“…To examine caspase-8 activity, we used the FAM-LETD-FMK FLICA (Carboxyfluorescein-LETD-FMK Fluorochrome-Labeled Inhibitor of Caspase) probe, which is cell permeable and binds specifically to active caspase-8 (47). Neutrophils were analyzed by ImageStream flow cytometry (Fig.…”

“…Moreover, there are attempts to apply therapies based on the combined action of anti-MUC1 and cisplatin complexes. Gornowicz et al revealed the anti-cancer action of platinum(II) complexes combined with anti-MUC1 in MDA-MB-231 and MCF-7 breast cancer cells [ 17 , 18 ].…”

“…Recent studies have shown the transient metal complexes, such as those containing copper and ruthenium, exhibited anti-tumor effects and pyrazolone can form various complexes with different transient metals to exhibit strong bio-activity 16 20 , but the cytotoxicity of pyrazolone-based copper complexes towards HeLa cells is not clear yet. The anti-tumor mechanism of pyrazolone based copper complexes remains poorly understood.…”
