Mechanisms of Activation of LRRC8 Volume Regulated Anion Channels

Abstract: Volume regulated anion channels (VRACs) are ubiquitously expressed in all vertebrate cells. Despite many years of research, the fundamental mechanisms underlying VRAC activation are not understood. The recent molecular identification of the LRRC8 genes underlying VRAC revealed that VRACs are formed by a hexameric assembly of members of the LRRC8 gene family. Knowing the genes underlying VRACs allowed the discovery of novel VRAC functions into cell volume regulation, and first structure function studies reveale… Show more

“…In our study, we found that açai berry restored autophagic flux at physiological levels. Interesting studies have shown that the altered expression of MAP-2 and β-tubulin leads to neuronal death; such results could be used in the future target for neuroprotective therapy targeting [ 111 , 112 , 113 , 114 ]. In our work, we found that açai berry stops the process of cell death and neurodegeneration and restores the microtubule network.…”

“…In physiological conditions, the production of reactive oxygen and nitrogen species generated during cellular metabolism in biological systems is balanced by the ability of the latter to defend through their sophisticated antioxidant machinery. Nevertheless, when oxidants are produced in excess, or when the antioxidant defenses that regulate them are ineffective, this balance can be perturbed, thus resulting in oxidative stress (OS) [ 4 , 7 , 8 , 9 , 10 ]. In these conditions, biomolecules can be altered through oxidation to an extent that exceeds repair capacity.…”

“…LRRC8A-E/SWELL1 proteins are essential components of the volume-regulated outwardly rectifying Cl – current (VRAC, VSOR, VSOAC, ICl swell , ICl vol ) elicited in most cells during cell swelling ( Qiu et al, 2014 ; Voss et al, 2014 ; Jentsch, 2016 ; Jentsch et al, 2016 ; Okada, 2020 ; Bertelli et al, 2021 ), and they are involved in a broad variety of cellular functions ( Chen et al, 2019 ). LRRC8A-E/SWELL1 also regulate the PI3K-Akt, Erk1/2, mTOR signaling cascade ( Kumar et al, 2020 ; Alghanem et al, 2021 ).…”
