Mechanisms of the immunosuppressive effects of mouse adipose tissue-derived mesenchymal stromal cells on mouse alloreactively stimulated spleen cells

Abstract: The mechanisms of immunomodulation by mesenchymal stromal cells remain poorly understood. In this study, the effects of mouse adipose tissue-derived mesenchymal stromal cells (ASCs) on mouse spleen cells alloreactively stimulated by anti-CD3 and anti-CD28 antibody-coated (anti-CD3/CD28) beads were observed. Production of interferon-γ by the anti-CD3/CD28 bead-stimulated spleen cells was significantly suppressed in co-culture with ASCs. However, an augmented intensity of CD69 on the stimulated spleen cells was … Show more

“…MSC were capable of inhibiting CD4 + cell activation (according to the decreased expression of CD69 and increased expression of CD62L on CD4 + cells in the presence of MSC, S1 Fig), i.e. exhibited characteristic immunosuppressive functions described in other studies [3032]. …”

“…These cells also protect against organ rejection and prevent from graft versus host disease after allogeneic stem cell transplantation (Yañez et al 2006). Immunomodulatory properties have been confirmed both in vitro and in vivo (Baer 2014; Le Blanc et al 2003; Nagaya et al 2014; Patel et al 2008). …”

“…AD-MSCs also express several kinds of growth factor receptors and by secretion of various cytokines and growth factors such as interleukin (IL)-6, IL-8, IL-10, HGF, VEGF, or FGF2, exhibit immunomodulatory, angiogenic, and antiapoptotic properties, in vitro or in vivo (Kilroy et al, 2007;De Francesco et al, 2015). AD-MSCs can act to suppress the proliferation and function of activated lymphocytes, inhibit the production of proinflammatory cytokines, and induce regulatory T cells (Patel et al, 2008;Gonzalez-Rey et al, 2010;Nagaya et al, 2014). AD-MSCs are also capable to reduce the damage, promote regeneration, or restore function of different tissues such as nervous system, heart, liver, or pancreas (Salgado et al, 2010;Zack-Williams et al, 2015;Bajek et al, 2016).…”
