Background Bulbus Fritillariae Cirrhosae (BFC) is an endangered high-altitude medicine and food homology plant with anti-tumor, anti-asthmatic, and antitussive activities as it contains a variety of active ingredients, especially steroidal alkaloids. Bulbus Fritillariae Thunbergia (BFT) is another species of Fritillaria that grows at lower altitude areas. Production of plant-derived active ingredients through a synthetic biology strategy is one of the current hot topics in biological research, which requires a complete understanding of the related molecular pathways. Our knowledge of the steroidal alkaloid biosynthesis in Fritillaria species is still very limited. Results To promote our understanding of these pathways, we performed non-target metabolomics and transcriptome analysis of BFC and BFT. Metabolomics analysis identified 1288 metabolites in BFC and BFT in total. Steroidal alkaloids, including the proposed active ingredients of Fritillaria species peimine, peimisine, peiminine, etc., were the most abundant alkaloids detected. Our metabolomics data also showed that the contents of the majority of the steroidal alkaloids in BFC were higher than in BFT. Further, our comparative transcriptome analyses between BFC and BFT identified differentially expressed gene sets among these species, which are potentially involved in the alkaloids biosynthesis of BFC. Conclusion These findings promote our understanding of the mechanism of steroidal alkaloids biosynthesis in Fritillaria species.

The severity and perseverance of the in ammation have been demonstrated in many health conditions.The limitations of existing medications, propose the need for newer alternative anti-in ammatory medications. In our earlier studies, we demonstrated the topical anti-in ammatory potential of crude ethanolic extract of Tetrastigma sulcatum leaves and its fractions. In the present study, we further explored anti-in ammatory activity of T. sulcatum extract, fractions, pure compound and its derivatives using in vitro and in vivo bioassay techniques. We attempted to isolate a pure compound from leaf extract and was identi ed as Friedelan-3β-ol (CI) and its derivatives Friedelinol acetate (C II) and Friedelinol methyl ether (C III) were synthesized. Treatment with crude extract and its fractions demonstrated a signi cant reduction in the mRNA expression levels of pro-inflammatory cytokines (IL-1β, IL-6, TNF-α) and nitric oxide (NO) production in LPS-stimulated in ammation in RAW 264.7 cells.Likewise, compounds CI, CII and CIII showed a similar pattern of signi cant inhibition of proin ammatory cytokines and NO production. In vivo study in Carrageenan induced paw-in ammatory mice model demonstrated reduced paw oedema and proin ammatory cytokines levels in a dose-dependent manner upon treatment of extract, its fractions, pure compound (CI), and their derivatives (CII and CIII.). The docking study showed all the compounds (CI, CII and CIII) share common residues with Dexamethasone. TNF-α exhibited the most interacting residues with the compounds. The present study con rmed the T. sulcatum 's anti-in ammatory activity, suggesting Friedelan-3β-ol as an active component in a crude extract.

Inflammation is a part of the intricate biological response of vascular tissues to undesirable stimuli such as infections, damaged cells, or irritants. Symptoms include redness, swollen joints, joint soreness, stiffness, and loss of joint function. Inflammation is now treated using non-steroidal anti-inflammatory drugs (NSAIDs). These drugs, however, increase the chance of blood clots, which may lead to heart attacks and strokes. As a consequence, the development of effective anti-inflammatory drugs based on natural substances is being studied. Natural products are a great source for the creation of innovative medications due to their chemical diversity. Many inflammation-related disorders may be treated with a natural product made from medicinal plants. The most often prescribed anti-inflammatory medicine has a slew of undesirable side effects. Because of these negative effects, researchers are looking for novel medications that have fewer or no side effects. Hundreds of phytoconstituents have been reported to have a range of pharmacological effects, however most of these researches are just of academic interest, and only a few make it into clinical trials. The purpose of this research is to assemble data on phytochemicals from herbal plants that have been tested in inflammatory models using cutting-edge scientific approaches.
