Meiotic differences between three triatomine species (Hemiptera, Reduviidae)

Abstract: We have found the following differences in the male meiosis among three triatomine species: (1) The three largest autosomal bivalents of Triatoma infestans are heterochromatic. Rhodnius prolixus has two autosoreal bivalents with heterochromatic blocks. Triatoma rubrovaria does not show any heteropycnotic autosomes. (2) Sex chromosomes in T. infestans form a chromocenter. At early prophase terminal associations are seen between sex chromosomes in T. rubrovaria, and they maintain a close association until diakin… Show more

“…For slide preparation, each tubule received two baths in distilled water for 5 minutes and then was placed in a 45% acetic acid solution for 10 minutes. The tubule was squashed and the material was stained with lacto-acetic orcein (De Vaio et al, 1985, with modifications according to Alevi et al, 2012a) and impregnated by silver ions (Howell and Black, 1980). The slides were examined under a Zeiss-Jenaval light microscope (magnified 1000X) and the images were captured using the Zeiss AxioVision LE 4.8 software (Carl Zeiss Microscopy GmbH, Jena, Germany).…”

“…After the seminiferous tubules were squashed, part of the material was stained with lacto-acetic orcein (De Vaio et al 1985) and part was submitted to silver nitrate staining (Howell and Black, 1980). Measurements of the sex chromosomes were performed with the Image Tool version 3 for Windows (UTHSCSA, 1995(UTHSCSA, /2002 in ten metaphase I cells.…”

“…The seminiferous tubules of adult males were torn and fixed to a cover slip. They then underwent the cytogenetic technique of Lacto-acetic orcein (De Vaio et al, 1985 with modifications according to Alevi et al, 2012) and analyzed using a Jenaval light microscope (Zeiss). For the characterization of the karyotype were analyzed 50 mitotic metaphases (I and II).…”
