Melanogenesis in normal and phenylthiourea‐treated eggs and larvae of Sarotherodon aureus

Abstract: The effects of phenylthiourea on the melanophores of eggs and larvae of Surotherodon nureus were investigated macroscopically and ultrastructurally. Depending on the precise time of application of the drug, partial or total inhibition of melanogenesis of the premelanosomes was obtained, with total inhibition producing artificial albinos. Coincidentally, phenylthiourea also appeared to suppress somatic growth and development of the fry, presumably due to an additional inhibitory effect on the thyroid gland.

“…In particular, zebrafish researchers use phenylthiourea to suppress pigmentation in developing embryos due to its ability to interfere with tyrosinase activity (25,26,34). Similarly, thiourea reduces pigmentation in coral trout grouper, tilapia, European eel, and Japanese flounder (33,35,36,27). Similar effects were also reported in zebrafish (31) treated with methimazole.…”

“…Such inhibitory effects of antithyroid drugs on larval growth and development have been reported in several fishes and cited as evidence for a role of thyroid hormones in early development of fish (Dales and Hoar 1954;Honma and Murakawa 1955;Lanzing 1984;Munro 1984). Phenylthiourea and thiocarbamides are known goitrogens inhibiting the iodination of tyrosine which is essential for the production of thyroid hormones (Mountcastle 1980;Ganong 1981).…”

“…oval bodies with the pigment melanin (Rogers et al 1997;Van Den Bossche et al 2006). The major function of melanosomes is to protect the DNA in the spinous-layer cells from harmful ultraviolet (UV) radiation (Lanzing 1984). Environmental factors and changes occurring within the body activate the melanophores, their increasing activity being manifested as intracellular responses.…”
