Melatonin added to freezing diluent improves canine (Bulldog) sperm cryosurvival

Abstract: Cryopreservation compromises sperm fertilising capacity due to a series of alterations in sperm structure and physiology; use of antioxidants such as melatonin, added to freezing media, may help to reduce sperm cryoinjury. To test the effect of melatonin on Bulldog [Canis lupus familiaris] sperm cryosurvival, spermatozoa were diluted in a standard freezing medium, cooled to 5°C, and added more freezing medium to obtain 200 x 106 cells/mL, and 5% glycerol. Diluted spermatozoa were added melatonin (0.0, 0.0005, … Show more

“…Unfortunately, the type of melatonin solvent was not reported in the study. In another study on Bulldogs [ 10 ], the authors tested the effect of melatonin in the freezing medium on sperm cryo-survival. Similar to us, they used DMSO as the melatonin solvent.…”

“…The protective effects of melatonin supplementation of the cryopreservation media on the porcine, bovine, ovine, equine and, human frozen-thawed spermatozoa have been previously reported in several independent studies [ 9 ]. Canine frozen-thawed ejaculated [ 10 ] or epididymal [ 11 ] spermatozoa have also benefited from in vitro supplementation of the melatonin in the freezing extender. However, multiple studies show that the extent of the protective effects is species-specific and highly dependent on the melatonin concentration.…”

“…Since its discovery as a component of cooling extender, egg yolk has been widely used in mammalian sperm cryopreservation to protect sperm from initial cold shock [ 12 , 13 ]. Moreover, many studies have verified the benefits of egg yolk-based extenders for canine semen cryopreservation [ 14 , 15 ]. However, it has been stated that the use of egg yolk interferes with sperm evaluation and the presence of particulate material in the extender may reduce fertility [ 16 , 17 ].…”

“…The addition of 1–2 mM of melatonin could also improve the sperm motility of canine sperm ( 17 ). In canine sperm, 0.002 and 0.0035 mol/L of melatonin received good results in the conservation of mitochondrial activity ( 16 ). The effect of melatonin on the acrosome in dogs is controversial.…”

“…On the contrary, the negative effect of excess melatonin concentration on spermatozoa has been described ( 15 ). Recently, several studies have observed the effect of melatonin supplementation on canine semen quality using several melatonin concentrations, e.g., 0.0005–0.0035 mol/L ( 16 ) and 0.5–2 mM ( 17 ). However, no reference to potential seasonal effects was made.…”
