Messenger RNAs bearing tRNA-like features exemplified by interferon alfa 5 mRNA

Abstract: The purpose of this work was to ascertain whether liver mRNA species share common structural features with hepatitis C virus (HCV) mRNA that allow them to support the RNase-P (pre-tRNA/processing enzyme) cleavage reaction in vitro. The presence of RNase-P competitive elements in the liver mRNA population was determined by means of biochemical techniques, and a set of sensitive mRNA species were identified through microarray screening. Cleavage specificity and substrate length requirement of around 200 nts, wer… Show more

“…We first identified, for the key factor considered (e.g., RNase P), a set of substrate mRNA species. This was done by testing viral and cellular mRNAs that share a common origin and others that do not, but that are found in the same cell type, as well as those that have either common or antagonistic activities . Then, we tested whether the identified mRNAs could be substrates for other RNA structure−dependent factors, or could interact with a different factor (for instance, UVC light) .…”

“…RNase P is the only specific factor that has been used to identify sensitive sites in the entire mRNA population of human cells (hepatocytes) . A set of competition assays on the standard RNase P reaction suggested the presence of a massive number of RNA structures recognized by RNase P within this mRNA population.…”
