Metabolomics: On the way to an integration of biochemistry, analytical chemistry, and informatics

Abstract: The review considers the development of a new science-metabolomics. The metabolome is mainly regarded as a set of primarily biochemical parameters, each of which or their ratio can serve as a potential biomarker that increases the sensitivity and/or specificity of diagnostics of diseases.

“…An ideal biomarker is characterized by a high sensitivity, specificity, and predictive value; it is reproduced in humans of different sexes and ethnic groups, and the procedure for its determination is cost-effective. However, rarely does such a marker stand alone as a single parameter; rather, it is a derivative of several original indicators [212,213]. At the same time, a complex of physiological and biochemical methods combined with an appropriate analytical platform should be relatively simple (low-invasive and noninvasive methods), universal (modular), and flexible (algorithmic).…”

“…It is important to emphasize that the methods of metabolomics are not intended to replace the methods of classical biochemistry (spectrophotometry, enzyme immunoassay, etc.). Metabolic profiling provides a unique opportunity to perform diagnosing and monitoring at a qualitatively new level only in conjunction with the methods of biochemical analysis [212].…”

“…Metabolomics using NMR methods give an efficient, with high accuracy and reproducibility screening platform that can classify new antimicrobial drugs by MoA, becoming an excellent complement to HTS evaluation, in this research field have been demonstrated using a metabolome model of Mycobacterium smegmatis the metabolomic chemical structure, biological activity and dose [43]. Finally, with the metabolomics profiles obtained is possible to predict the toxicity response in vitro and in vivo of a specific molecular target or a drug formulation [44].…”

“…[14][15][16][17][18][19]. At the same time, for practical purposes, it seems promising to search for simple informative indices, for example, based on the ratio of indicators that are in antiphase over a close time interval [20][21][22][23]. For example, among the well-known indices are the De Ritis ratio, the index of atherogenicity, and the body mass index.…”
