Microarray-based detection of Korean-specific BRCA1 mutations

Abstract: A reliable multiplex assay procedure to detect human genetic mutations in the breast cancer susceptibility gene BRCA1 using zip-code microarrays and single base extension (SBE) reactions is described. Multiplex PCR amplification was performed to amplify the genomic regions containing the mutation sites. The PCR products were then employed as templates in subsequent multiplex SBE reactions using bifunctional primers carrying a unique complementary zip sequence in addition to a mutation-site-specific sequence. “…To show that two-step PCR is possible with SD polymerase, we used human genomic DNA as the template and a primer set that amplified a BRCA -related gene (Online Resource Table 1) [ 5 ]. To compare the sensitivity of the three-step qPCR and two-step qPCR processes, serial dilutions of the DNA template with 300, 30, 3, and 0 copies were tested in duplicate assays .…”

“…DNA targets are mainly used for cancer diagnosis, such as the BRCA1 and BRCA2 genes, which are associated with breast, ovarian, pancreatic, and prostate cancers [ 3 , 4 ]. Accurate BRCA testing is essential for early detection, identifying gene mutations, and selection of treatments [ 5 , 6 ]. RNA viruses include Zika [ 7 ], Ebola [ 8 ], and influenza [ 9 , 10 ], as well as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) [ 11 ], first identified in December 2019.…”

“…The invention of the PCR technique has made a tremendous impact on the areas of biological research and diagnostics since it can be used to generate quantities of nucleic acids needed for detection and measurements. However, the need for a temperature cycling instrument limits applications of PCR in point-of-care testing (POCT) environments . In order to overcome this limitation, several isothermal nucleic acid amplification methods have been developed.…”
