Background: Candidiasis is a spectrum of opportunistic fungal diseases that can manifest in drug addicts in various forms, such as stomatitis. Objectives: This study aimed to determine the prevalence of oral candidiasis caused by Candida glabrata in addicts and examine the susceptibility of isolates to two azoles. Patients and Methods: After taking oral samples from 131 drug addicts suspected of having oral candidiasis, Candida species were identified by culture on chromogenic Candida agar, carbohydrate-assimilation (API test), and polymerase chain reaction. The minimum inhibitory concentrations (MICs) of fluconazole and voriconazole against the isolates were determined using the broth microdilution method. Data were analyzed by SPSS (version 23) using the t-test and one-way analysis of variance. A P value of less than 0.05 was considered statistically significant. Results: Overall, 22.2% of the isolates were C. glabrata, which was more abundant among opiate addicts. Among these isolates, 61.1% were resistant to fluconazole and 44.4% were resistant to voriconazole. The MIC90 of voriconazole was 8 µg/mL, which was 16 times less than that of fluconazole (128 µg/mL). There was no statistically significant difference between the frequency of fluconazole-and voriconazole-resistant isolates (P > 0.05). Conclusions: Oral candidiasis was a common problem among drug addicts. In addition, the prevalence of azole-resistant C. glabrata isolates was high among these individuals.

Background and Objective: The prevalence of the infections caused by Candida species has led to a significant increase in their resistance to antifungal compounds. The aim of this study was to i) investigate drug resistance ii) evaluate the incidence of Candida albicans drug resistance pattern in the vaginal samples of women referring to health centers of Qom province, and iii) examine the effect of Zn nanoparticles combined with fluconazole against C. albicans isolates. Methods: This experimental, descriptive study was performed on 120 patients of candidiasis. In order to identify Candida albicans, direct experiments, differential culture, Germ tube test and sugar assimilation test (API20C kit) were conducted. The effect of different antifungal drugs and zinc nanoparticles and the synergistic effect of fluconazole with zinc nanoparticles were investigated by disk diffusion method. Minimum Inhibitory Concentration (MICs) of all cases was further specified. Results: Of the 120 samples, 50 (41.6%) were identified as Candida albicans. These strains were resistant to certain antifungal drugs while others were semi-sensitive and sensitive. The lowest and the highest mean diameter of inhibition zone in all Candida albicans isolates belonged to ketoconazole (15.64 mm) and fluconazole nano-ZnO (26.76mm), respectively. The lowest and the highest MICs were observed in fluconazole-nano-ZnO and nano-ZnO, respectively. Conclusion: The synergistic effect of Zn nanoparticles with fluconazole can be conducive to the treatment of vaginal candidiasis.

Background: Vaginal candidiasis is one of the most common vaginal infections in women of reproductive age group caused by Candida species of fungi, a normal inhabitant of vagina. Aims: The aim is to study the culture characteristics, species prevalence, and drug sensitivity of vulvovaginal candidiasis (VVC) among females with genital pruritus and discharge per vaginum who are proved to have Candida infection by a vaginal smear study. Materials and Methods: A prospective study was done on female patients aged 18 years attending the Sexually Transmitted Diseases Clinic of the Department of Dermato-Venereo-Leprology (DVL), Tirunelveli Medical College, with complaints of genital pruritus and vaginal discharge over a period of 1½ years from 2013 to 2015. After clinical examination, vaginal discharge was smeared for wet mount, potassium hydroxide mount, and Gram stain. Those with pseudohyphae were chosen for culture, speciation, and drug sensitivity. Results: The most common age group affected was 18–35 years. The most common predisposing factor was HIV infection-induced immunosuppression (48.7%), followed by antibiotic usage and diabetes mellitus. Clinical presentation of VVC was similar in HIV-positive and HIV-negative patients. Recurrent VVC makes up 12% of total cases with most common species being Candida albicans whereas Candida glabrata among the non- C. albicans . Drug sensitivity pattern of C. albicans showed the highest sensitivity to nystatin 85.7%, followed by triazoles (75%–85%) with ketoconazole being more resistant (40%–60%), and was similar in C. albicans and non- C. albicans . Limitations: Exclusion of unmarried and pregnant women and lack of follow-up were limitations in our study. Conclusions: According to our study, elimination of predisposing factors, speciation of fungus, and treatment based on drug sensitivity pattern will reduce the incidence of VVC.
