Microfluidic device with integrated microfilter of conical-shaped holes for high efficiency and high purity capture of circulating tumor cells

Abstract: Capture of circulating tumor cells (CTCs) from peripheral blood of cancer patients has major implications for metastatic detection and therapy analyses. Here we demonstrated a microfluidic device for high efficiency and high purity capture of CTCs. The key novelty of this approach lies on the integration of a microfilter with conical-shaped holes and a micro-injector with cross-flow components for size dependent capture of tumor cells without significant retention of non-tumor cells. Under conditions of consta… Show more

“…Modern filter membranes have utilized lithographic methods to precisely pattern pores into silicon 193,194 and polymers such as parylene-C, 190,195,196 poly(ethylene glycol diacrylate) (PEGDA), 197 and PDMS. 198 However, the cost for filter fabrication has ranged from extremely expensive palladium microfilters 199 to multi-stage deep reactive ion etching (DRIE) for each set of silicon or parylene-C microfilters 190,193 and replication of a lithographically-patterned master by PEGDA photo-polymerization 197 or PDMS casting.…”

“…Using size information, filtration method is fast, simple, straightforward, and reliable. [49][50][51][52][53] Recently, a filtration chip with lateral flow was combined with the vertical flow into the filter to capture the CTCs gently ( Fig. 2(a)).…”

“…3 Enumeration of CTCs in the cancer patient peripheral blood is useful for the prognosis, prediction of response to therapy and guiding the clinical decision after therapy. 4 Various methods have been developed to capture circulating tumor cells (CTCs), such as size or antibody-based microfluidics chips, [5][6][7][8][9] nano-micro interface based substrates. [10][11][12][13] Circulating tumor cell clusters in the blood are more important because they are oligoclonal precursors of cancer metastasis, such as breast cancer 14 .…”
