-The objective of this work was to evaluate the large-scale propagation of grapevine genotypes after short-term storage in vitro. Microshoots from ten grapevine genotypes were used. The following storage temperatures were evaluated: 10, 20, and 25°C. After short-term storage, the shoots were propagated in up to five successive subcultures, to assess the large-scale propagation of the germplasm maintained under conditions of minimal growth. The propagated shoots were rooted in different concentrations of indolbutiric acid (IBA) and acclimatized in greenhouse. The best temperature for short-term storage in vitro and survival of the genotypes was 20°C. In the propagation phase, the highest number of shoots per explant was found in the subcultures 4 and 5, with averages of 4.9 and 4.8 shoots per explant, respectively. In the rooting phase, the best results for number of roots were obtained using a culture medium supplemented with 0.4 µmol L -1 of IBA, with an average of three roots per shoot. During the acclimation phase, a survival rate higher than 95% was achieved after 30 days in the greenhouse. Grapevine genotypes maintained for six months in vitro, at 20 o C, can be micropropagated in large scale.

Dipteryx alata Vog. (barueiro) is a native fruit of the Cerrado used in food and medicine. The species has never been domesticated and is currently at risk of extinction. Plant tissue culture is an important tool for the conservation of germplasm and for quick, large-scale propagation of quality seedlings. While, this technique has never been used with barueiro, it could be a valuable tool in the domestication of this species. The objective of this work is to evaluate concentrations of MS medium salts (25, 50, 75 and 100%) supplemented with coconut water (100 mL L -1 ), dwarf banana pulp (60 g L -1 ) or the absence of any supplement in the in vitro establishment of barueiro seeds (Dipteryx alata Vog). The experiment used a completely randomized design (CRD) with a 3x4 factorial scheme (three cultivation media types and four concentrations of MS salts) and three repetitions. The following characteristics were evaluated: germination rate, shoot length , largest root length , stem diameter , number of leaves , chlorophyll A , chlorophyll B , total chlorophyll, and the fresh and dry mass of the seedlings. The coconut water and banana pulp supplements did not improve the in vitro development of the barueiro seedlings and the full concentration of MS salts did not improve the initial development of the seedlings. However, the pure MS medium at a concentration of 25% MS salts was the best option for the in vitro establishment of Dipteyx alata.

A faveleira (Cnidoscolus phyllacanthus Pax et K. Hoffm.) é uma xerófita forrageira do semi-árido do Nordeste do Brasil. A presença de espinhos causticantes dificulta a coleta de suas ramas, no entatnto mutantes inermes podem ser multiplicados vegetativamente. Este trabalho teve o objetivo de determinar um protocolo de enxertia por garfagem da faveleira sem espinhos com enxertos dormentes ou em fase vegetativa enxertados em porta-enxertos na fase vegetativa. As enxertias foram realizadas no Viveiro Florestal da UFCG, campus de Patos, PB, em dezembro de 2003 e junho de 2004. Antes da enxertia, em dezembro de 2003, os enxertos dormentes foram mantidos por três dias sob refrigeração (+5°C) (T1) ou à sombra, à temperatura ambiente (T2). Os enxertos em fase vegetativa foram enxertados logo após cortados, em dezembro de 2003 (T3) ou junho de 2004 (T4). Foram realizadas 38, 26, 40 e 30 enxertias para T1, T2, T3 e T4, respectivamente. Nos enxertos que brotaram, foram avaliados o diâmetro e a altura do porta-enxerto e do enxerto, e o percentual de sucesso das enxertias. As médias de diâmetro (10,9 a 11,9mm e 8,6 a 9,6mm dos porta-enxertos e enxertos, respectivamente) e altura (7,7 a 12,6cm e 23,7 a 29,8cm dos porta-enxerto e enxertos, respectivamente) foram considerados semelhantes para todos os tratamentos. Os percentuais de sucesso das enxertias para T1, T2, T3 e T4 foram 45, 85, 15 e 33%, respectivamente. Concluiu-se que enxertos dormentes de faveleira, com cerca de 9mm e 25cm de altura, e mantidos à sombra por três dias antes de serem enxertados, propiciaram a maior taxa de sucesso das enxertias.
