MicroRNA‑206 relieves irradiation‑induced neuroinflammation by regulating connexin 43

Abstract: Radiation therapy has been widely used for the treatment of various types of cancer; however, it may cause neuroinflammation during the pathological process of the disease. Astrocytes, the most abundant cell type in the central nervous system, have been confirmed to play vital roles in various diseases. Connexin (Cx)43, the main Cx type in astrocytes, which has been identified as a direct target gene of microRNA (miR)-206, was found to be involved in diseases pathologies in regions with astrocytes. The aim of … Show more

“…Due to their critical functions, connexin expression and channel activity are tightly regulated by various factors, including extracellular Ca 2+ , intracellular pH, membrane potential, post-translational modifications, and epigenetic regulation (Aasen et al, 2018;. Moreover, non-coding RNAs including microRNAs can also regulate connexin expression (Curcio et al, 2013;Zeng et al, 2021). Extracellular Ca 2+ exerts a significant influence on connexin channel gating by directly binding to the channels.…”

“…In cultured γ-radiation-induced astrocytes, miR-206 significantly lowered Cx43 expression and inflammatory cytokine release. In contrast, the Cx43-plasmid had the opposite effect, revealing that miR-206 relieves neuroinflammation by downregulating Cx43 [ 34 ]. Astrocytes transfected with miR-301a-3p, which can bind to Cx43 and downregulate its expression, have exhibited increased proliferative potential and decreased apoptosis; however, astrocytes overexpressing the Cx43 protein have exhibited high cell death rates [ 35 ].…”

“…All the research presented supports placing Cx and Panx in the center of the discussion about neuroinflammation and provides evidence to encourage the study of new pharmacological agents that aim specifically at these cellular structures when treating inflammationHigh yield treatments for neuroinflammation are raising in all areas of study, as seen in new treatments for neuroinflammation caused by irradiation (prevalent in cancer patients), innovative gene therapy may be upcoming in the following years as, Wei Zegn. Et al research with in vitro cells showed that microRNA-206 can relieve irradiation induced neuroinflammation, this was the case of transfected cells with miR-206 that not only reduced the inflammatory profile but also reduced Cx43 expression despite of being exposed to radiation [ 194 ].…”
