MicroRNA-338-3p inhibits glucocorticoid-induced osteoclast formation through RANKL targeting

Abstract: ABSTRACT. The differentiation deficiencies of osteoclast precursors (pre-OCs) may contribute to osteoporosis. Research on osteoporosis has recently focused on microRNAs (miRNAs) that play crucial roles in pre-OC differentiation. In the current study, we aimed to analyze the expression and function of the glucocorticoid (GC)-associated miRNA-338-3p (miR-338-3p) in osteoclast formation. We found that dexamethasone induced osteoclast differentiation and inhibited miR-338-3p expression. Overexpression of an miR-33… Show more

“…Along with their known micro‐RNA interactions, these lncRNAs may hold potential as future experimental targets. For instance, lncRNA LINC01225 , which was strongly positively correlated with RANKL and also 2.64‐fold higher in peri‐implantitis, is predicted to interact with miR‐338‐3p suggesting possible therapeutic value of their specific modulation.…”

“…For instance, osteoclasts could secrete miRNA-21 that promoted tumor cell proliferation 17 and miRNA-378 that enhanced tumor growth, angiogenesis and cell survival 18 . Also, miRNA-338-3p was demonstrated to repress glucocorticoid-induced osteoclast differentiation and bone resorption through targeting on receptor activator for nuclear factor-κB ligand (RANKL) 19 .…”

“…Pituitary MicroR-449a (miR-449a) impairment induced a decrease in the level of CRH receptor 1 (CRHR1) level in low-birth-weight rats (Nemoto et al, 2015 ), and miR-449a was involved in the GC induced CRHR1 downregulation (Nemoto et al, 2013 ). GC function was related to miR-338 (Zhang et al, 2016 ) and miR-433 (Smith et al, 2016 ) in osteoclast formation. Hypothalamic miRNAs reportedly played a critical role in energy balance (Schneeberger et al, 2015 ).…”
