MicroRNA-494 Targeting Both Proapoptotic and Antiapoptotic Proteins Protects Against Ischemia/Reperfusion-Induced Cardiac Injury

Abstract: Background-MicroRNAs (miRs) participate in many cardiac pathophysiological processes, including ischemia/reperfusion (I/R)-induced cardiac injury. Recently, we and others observed that miR-494 was downregulated in murine I/R-injured and human infarcted hearts. However, the functional consequence of miR-494 in response to I/R remains unknown. Methods and Results-We generated a mouse model with cardiac-specific overexpression of miR-494. Transgenic hearts and wild-type hearts from multiple lines were subjected t… Show more

“…Importantly, TNFα, which increases upon cellular rejection 34, upregulates miR‐494 35 and downregulates miR‐149‐5p 36, in line with our findings. The upregulation of miR‐494 might be beneficial as it prevents ischemia‐reperfusion injury 37. The downregulation of miR‐149‐5p may contribute to the induction of genes downstream of the proinflammatory cytokine IL‐6 38, which we also observed in our mRNA arrays.…”

“…MiR-494 is known as an oncomir in gastrointestinal stromal tumors targeting the proto-oncogene KIT (28). In addition, miR-494 enhances myocyte survival by targeting PTEN, ROCK1, and CAMKIId and protects against ischemia/reperfusion-induced cardiac injury (29). miR-494 is also an essential player in regulating the accumulation and activity of myeloidderived suppressor cells by targeting PTEN and activation of the Akt pathway (25).…”

“…And miR-21 regulates fibroblast metalloprotease-2 in murine myocardial infarction 31 . Also, miR-494 targets to PTEN, ROCK1 and CaMKIId in the cardiac I/R injury 32 . In the future, we will unveil more regulators that function in I/R response and provide more valuable therapeutic targets for effective treatment of cardiac diseases.…”
