MicroRNAs and their role in the malignant transformation of oral leukoplakia: a scoping review

Abstract: Background MiRNAs are small non-coding RNAs that regulate gene expression at the post-transcriptional level and have been associated with malignant transformation of oral epithelial precursor lesions such as oral leukoplakia. The aim was to perform a scoping review of the contemporary literature about the different roles of miRNAs during the malignant transformation of oral leukoplakia. Material and Methods We conducted a systematic search with the following MeSH terms:… Show more

“…The study of miRNA holds promise. Different systematic reviews have also concluded that they are potential biomarkers for OSCC and OPMDs [ 11 , 72 , 73 ]. The main issue is, as with many of the other markers, is that apart from mir-21, which was assessed by two studies [ 47 , 48 ], the salivary expression of all other miRNAs, has been analyzed by single studies.…”

“…Saliva is probably the most attractive secretion for the finding of biomarkers for OSCC and OPMDs, as its collection is easy, non-invasive, fast, and cost-effective. Thus, different studies have explored the possibility of using salivary factors as biomarkers for the development and progression of OSCC and OPMDs [ 9 , 10 , 11 ]. The aim of this study was to perform an exploratory systematic review about the different salivary molecules that have been assessed as possible biomarkers for the diagnosis of OSCC.…”

“…It is worth noting that the differentially expressed miRNAs in OLK are identified to associate with its malignant transformation [ 63 66 ]. In the latest scoping review, the authors have included fifteen articles for analysis and identified 21 differentially expressed miRNAs in OLK [ 67 ]. To be specific, 6 miRNAs (miR-21, miR-181b, miR-184, miR-345, miR-146a and miR-208-3p) that were categories as oncogenic, 5 miRNAs (miR-31*, miR-3065-5p, miR-129-2-3p, miR-204-5p and miR-375) that were classified as tumor suppressors and 10 miRNAs (miR-138, miR-639, miR-27a-3p, miR-15b, miR-485-5p, miR-200c, miR-19a, miR-424, miR-205 and miR-21) that were related to epithelial–mesenchymal transition, invasion and migration [ 67 ].…”

Among these miRNAs, the authors concluded that miR-21, miR-345, miR-181b, miR-31* might be potential markers of malignant transformation of OLK.…”

“…miRNA-375 was shown to be downregulated in OSCC and progressive oral leukoplakia compared to non-progressive lesions, indicating that it functions as a tumor suppressor by decreasing SLC7A11 expression during oral carcinogenesis [12]. Specific miRNAs can cause cancer cells to undergo ferroptosis by decreasing SLC7A11 expression [13].…”
