Microscopy and Image Analysis

Abstract: This unit provides an overview of light microscopy, including objectives, light sources, filters, film, and color photography for fluorescence microscopy and fluorescence in situ hybridization (FISH). We believe there are excellent opportunities for cytogeneticists, pathologists, and other biomedical readers, to take advantage of specimen optical clearing techniques and expansion microscopy-we briefly point to these new opportunities. © 2017 by John Wiley & Sons, Inc.

“…Objectives also naturally present with color artifacts, that is, chromatic aberrations or color “fringes.” Objectives that have been color corrected via the combination of unique glasses, each holding different color refraction properties, are followed by the suffix “apochromat.” Historically, apochromatic lenses would correct for three to four spectral lines. [ 71 ] Some modern WSI models include apochromatic lenses correcting for up to 14 spectral lines, that is, fully color corrected, for example, Carl Zeiss apochromatic.…”

“…Chromosome analysis may be done through the microscope or by the use of digital images with karyotyping analysis software on desktop computers. A number of digital‐image‐capture devices are available ( UNIT , McNamara et al., ). These devices, which can substantially speed the counting and analysis of chromosomes and provide a digital “photograph” of chromosomes, are currently in use in most cytogenetic laboratories.…”

“…Digital image analysis method (Calvaruso et al, 2009; Chen, Li, Dong, & Pei, 2018; Madabhushi & Lee, 2016; McNamara, Difilippantonio, Ried, & Bieber, 2017) can be used to perform quantitative evaluations of reticular fiber fragments. Fragment counting under a microscope can be carried out by a specialist by continually looking at the microscope lens or by automatic cell counting methods (Belsare & Mushrif, 2012; Gurcan et al, 2009; Madabhushi & Lee, 2016;Yegin et al, 2015; Yegin, Yegin, & Ozdogan, 2016).…”
