The aim of the present study was to characterise milk of Burlina local cattle breed for traits of technological and nutritional relevance, such as milk coagulation properties (MCP), and protein, major mineral and fatty acid (FA) composition. Burlina is mainly reared in mountain areas of Veneto Region (Italy) and it has been inserted in conservation plans aiming to avoid biodiversity loss and marginal pasture areas abandonment. Eighty-one individual milk samples were collected in four farms. Milk coagulation properties were determined using Formagraph, and protein, mineral and FA composition were analysed in high performance liquid chromatography, inductively coupled plasma optical emission spectrometry and gas chromatography, respectively. Results evidenced good protein percentage (3.38%) and considerable casein content (28.89 mg/mL), as well as a desirable FA profile, with x-6 to x-3 ratio of 4.04. Somatic cell score, averaging 3.13, is a trait that should be enhanced through the improvement of farm management. This would have positive effects on MCP. Among milk minerals, the most and less abundant were K (1493.53 mg/kg) and Mg (110.07 mg/kg), respectively. Overall, herd, parity and lactation stage explained moderate to low variation of the studied traits. Results of the present study could be useful to valorise Burlina local breed and preserve biodiversity in marginal areas. ARTICLE HISTORY

Abstract. Raman spectroscopy has become a powerful and popular tool for food systems analyses lately. Based on characteristic vibrations of the studied material, the information on its content and structure can be answered. In the paper, Raman spectroscopy is studied for a purpose of lactose content in milk assessment. Lactose, the milk disaccharide, in a human organism decomposes during digestion by the act of enzyme lactase to more easily digestible monosaccharides -glucose and galactose. The lack of enzyme lactase causes symptoms of lactose intolerance what limits lactose-intolerant individuals in the intake of milk and dairy products. Lactose-free products in the diet can be a solution. Raman spectroscopy offers rapid measurement independent of the number of chemicals and other in the paper listed benefits. Raman spectra of lactose, glucose and galactose exhibit enough differences to distinguish the content of lactose in milk. C-O-H bending mode at 1087 cm -1 is used for lactose quantification. The method accuracy for measuring content of lactose was tested on dried milk droplets.Evaluation of the spectroscopic data was related to two different substances -phenylalanine contained generally in the milk and crystal violet used as an internal standard.

Adulteration of goat milk is usually done using cow's milk product. Cow milk is used as it is widely available and its price is cheaper compared to goat milk. This paper shows a development of candidate tools for milk adulteration using cow milk. A quartz crystal microbalance immunosensor was developed using commercial crystal resonator and polyclonal antibody specific to cow milk protein. A specific protein at 208 KDa is found only in cow milk and does not exist in goat milk. The existence of this protein can be used as an indicator of cow milk content in a target solution. To detect the PSS 208 kDa protein, antibody specific to the PSS 208 was developed. The purified antibody was immobilized on top of the sensor surface on a polystyrene layer. The fraction of the immobilized antibody on the sensor was found at 1.5% of the given antibody. Using a static reaction cell, the developed immunosensor could detect the specific cow milk protein in buffer solution. The detection limit is 1 ppm. A linear relationship between frequency change and specific protein of cow milk concentration is found from a concentration of 1 ppm to 120 ppm.
