MiR-140 inhibits classical swine fever virus replication by targeting Rab25 in swine umbilical vein endothelial cells

Abstract: Classical swine fever virus (CSFV) is one of the most important viral pathogens leading worldwide threats to pig industry. MicroRNAs (miRNAs) play important roles in regulating virus replication, but whether miRNAs affect CSFV infection is still poorly understood. In previous study, we identified four miRNAs that were down-regulated by CSFV in swine umbilical vein endothelial cells (SUVEC). In this study, miR-140, one of the most potently down-regulated genes was investigated. We found that the miRNA expressio… Show more

“…miRNA is closely related with viral infection. Viral infection alters host miRNA expression, and in turn, host miRNA regulates viral infection (Trobaugh and Klimstra 2017;Lin et al 2020;Wang et al 2019;Xu et al 2020). Most multicellular organisms and several viruses can generate miRNAs (Andreassen and Høyheim 2017a;Gutkoska et al 2017;Liu et al 2017b;Wang and Zhu 2017;Yang et al 2017a;Yu et al 2017;Huang et al 2019;Yoon et al 2020;Zhao et al 2020).…”

“…The wild-type (WT) plasmids of circ_0016760 (circ_0016760-WT) and HIF1A (HIF1A-WT) and their mutant (MUT) plasmids (circ_0016760-MUT and HIF1A-MUT) were constructed by Ribobio Co., Ltd. Cell transfection was performed according to the literature’s methods [ 25 ]. Luciferase activities were detected using a Dual-Lucy Assay Kit (Solarbio).…”

“…These host proteins could be divided into the following categories according to their possible mechanism involved in CSFV replication. (1) Many members of the small Rab GTPase family, such as Rab1A [58], Rab1b [59], Rab2 [60], Rab5 [61,62], Rab7, Rab11 [62], Rab18 [63], and Rab25 [64], play a critical role in CSFV replication. For example, Rab18 interacts with CSFV NS5A and mediates viral RNA replication and virion assembly [63].…”

“…MiRNAs could also regulate CSFV replication. For example, miR-140 inhibits CSFV replication by targeting Rab25 [64]. However, there are still few studies on the relationship between miRNA and CSFV replication.…”
