MITOSIS IN THE YEAST LIPOMYCES LIPOFER

Abstract: A study of mitosis in Lipomyces has been carried out because preliminary observations by Ganesan and Roberts, 1959 (9), had indicated that the nucleus of this yeast might be unusually favourable for morphological observations. This impression has proved correct. The chromosomes of Lipomyces are visible as separate, countable bodies for the greater part of mitosis. The pattern of mitosis differs from the common one in that in Lipomyces the proper distribution of sister chromosomes is accomplished without the hel… Show more

“…However, imperfect yeasts cannot be studied using traditional genetics. Another approach to obtain direct evidence for chromosome numbers is cytochemical staining of mitotic chromosomes with Giemsa, e.g., in Kluyveromyces lactis and Saccharomyces cerevisiae (Galeotti & Williams 1978), Lipomyces lipofer (Robinow 1961), andSchizosaccharomyces pombe (Fisher et al 1975). DNA-binding fluorochromes like DAPI, mithramicin and para-rosaniline do not reveal distinct chromosomal patterns at the light microscopic level.…”

“…Thus, the immunofluorescent staining pattern of Ndcl0p is for the most part consistent with the proposed pattern of kinetochore localization, but there are some inconsistencies and other possible interpretations of the staining pattern. Some of these problems may be resolved by study of a yeast with more favorable chromosome morphology, for example Lipomyces lipofer (Robinow, 1961).…”

“…This organism belonging to another family (Saccharomycetaceae) of the Endomycetales has been thoroughly studied by Robinow (1961). According to Robinow the nucleus is small and compact in the full grown bud or detached single cell.…”

“…In Lipomyces the 'stack' divides transversely, wheras in N. coryli the two elongated nuclei move along each other in opposite directions. Matile, Moor & Robinow (1969) state: 'Seeing the chromosomes, however, is not the same as understanding their behaviour, and the course of mitosis in Lipomyces lipofer (Robinow, 1961) needs reinvestigating'. Also from a genetic viewpoint it is difficult to liken a transverse constriction to the division of an elongated nucleus.…”
