Mixed Solvent Method for Improving the Size Uniformity and Cargo-Loading Efficiency of ZIF-8 Nanoparticles

Abstract: Zeolitic imidazolate framework-8 (ZIF-8) nanoparticles with tunable diameters and a uniform morphology were constructed in dimethyl sulfoxide (DMSO)/H 2 O mixed solvents and were further decorated with dextran to improve their stability and biocompatibility. A series of reaction conditions, including the DMSO content in mixed solvents, molar ratio between precursors, growth time, and decoration of dextran, were systematically investigated. Most importantly, it was the union of DMSO and water that achieved the … Show more

“…Meanwhile, in the water/DMSO system, more DTX was encapsulated than in methanol or water. 52 The above results show that DTX can be efficiently loaded into the nanoparticles. Furthermore, the stability of DTX@ZIF-8 and RVG 15 -PEG@DTX@ZIF-8 in water was evaluated after storage for 7 d at 4 °C.…”

“…DTX@ZIF-8 nanoparticles were prepared using a simple one-pot synthesis as described in previous reports, with some modifications. 52 In detail, 5 mg of DTX (Meilun, China) and 80 mg of Zn(NO 3 ) 2 ·6H 2 O (Macklin, China) were dissolved in 3 mL of DMSO. The mixture was then stirred for 30 min.…”

“…This detection strategy relied on the incubation of cell lines with fluorescent ZIF-8 (FZIF-8) nanoparticles and the subsequent flow cytometry analysis of these cells based on the hypothesis that the FZIF-8 nanoparticles would display a specific fluorescent response in DR cancer cells that could be readily distinguished from that in DS cancer cells (Scheme C). Briefly, the FZIF-8 nanoparticles were prepared via the method described in our previous work, , and they exhibited a polyhedral morphology with a diameter of ∼123 nm (Figure S1A,B). The hydrodynamic diameter measured by dynamic light scattering (DLS) was about 143 nm (size polydispersity index (PDI) = 0.212), which reflects the excellent dispersity of FZIF-8 nanoparticles in water (Figure S1C).…”

“…Nonetheless, ZIF‐8 synthesized in aqueous systems typically exhibits a lower surface area compared to those prepared in organic systems. In 2021, Guo et al [ 30 ] successfully synthesized ZIF‐8 with exceptional uniformity and regularity using a solvent mixture of dimethyl sulfoxide (DMSO) and water. Furthermore, they decorated these nanoparticles with dextran so as to improve chemical stability and biocompatibility.…”
